高效液相色谱测定紧压茶中多种有机酸条件研究

【摘要】 建立了一种利用高效液相色谱测定紧压荼中10种有机酸的方法.紧压荼中有机酸的色谱条件为:Diam onsil C18柱(5i,250×4.6mm),柱温30℃,流速0.6 ml/min,检测波长 210nm,进样量15i 1,流动相0.1 mol/L KH2PO4(用磷酸调至pH 2.8).该方法有机酸添加回收率达到92.5%-101.3%,相对标准差0.62%-2.73%,各种有机酸浓度与HPLC峰面积的线性相关系数大于0.94,表明该测定方法快速简便,具有较高的准确度和精确度.

【Abstract】 The determination of ten organic acids in compressed tea by using high performance liquid chromatography was investigated. Organic acids were analysed by using a Shimadzu LC-2010 HPLC equipped with a UV detector and a Diamonsil C18 column, 15-i 1 sample was injected into the column. The detection was performed by measuring absorbance at 210 nm and column temperature at 30℃. The flow rate was set at 0.6 ml/min and mobile phase was 0.1mol/L KH2PO4 (pH value was adjusted to 2.8 by phosphoric acid). The recoveries of the spiked organic acids and the relative standard deviations of the organic acids were 92.5% - 101.3%, and 0.62%-2.73%, respectively. All of the correlation coefficients between the concentration of the organic acids and the HPLC response areas were higher than 0.94, indicating that the method was simple, rapid and accurate.
