牛膝多糖对老龄大鼠非特异性免疫功能影响作用研究

【摘要】 目的探讨牛膝多糖(ABPS)对老龄大鼠非特异性免疫功能的调节作用.方法牛膝多糖(ABPS)在老龄大鼠背部皮下注射21天,镜检计外周血白细胞数(WBC),HiCN法测定血红蛋白(Hb)含量,用全自动生化分析仪测定主要生化指标,用含10%小牛血清的RPMI-1640细胞培养液贴壁培养肺泡巨噬细胞(AMΦ)和腹腔巨噬细胞(PMΦ)2h,并用全自动生化分析仪测定其乳酸脱氢酶(LDH)和酸性磷酸酶(ACP)活性,摄取中性红试验检测巨噬细胞(MΦ)的吞噬功能,比色法检测脾脏、肝脏、大脑、肾脏和血清中的脂质过氧化物(LPO)和还原型谷胱甘肽(GSH) 含量.结果牛膝多糖(ABPS)能显著提高老龄大鼠AΦ中和PMΦ内ACP和LDH活性(P<0.05;P <0.05),显著增强AMΦ和PMΦ摄取中性红的能力(P<0.001);且肝脏、大脑中的LPO含量下降,GSH含量上升.结论牛膝多耱(ABPS)具有激活老龄大鼠AMΦ和PMΦ作用,能增强其机体的非特异性免疫功能.

【Abstract】 Objective To study regulation of Achyranthes bidentata polysaccharide (ABPS) on non-specificity immune function of old rates. Methods After subcutaneous injected with ABPS (50mg/kg weight) in the back of the old rats for 21 days, the WBC and the levels of Hb and chief Biochemical index in blood of old rats were determined; After the alveolar macrophages( AM Φ) and peritoneal macrophages (PM Φ)cultured with 10% Fetal Bovine Serum-RPMI-1640 for 2h in vitro, the activities of LDH and ACP in the AM Φ and PM Φ were detected by automatic biochemistry analyzer, and the abilities in devouring neutral red in the AM Φ and PM Φ were observed. The levels of LPO and GSH in some immune organs like spleen, liver, brain, kidney and in serum were also detected. Results The levels of ACP and LDH in the AM Φ or PM Φ were increased(P<0.05;P<0.05), the ability of AM Φ or PM Φ in devouring neutral red was increased (P<0.001), the levels of LPO were diminished and the levels of GSH raised in the liver and brain. Conclusion ABPS has the functions of activating the AM Φ and PM Φ, it can heighten non-specificity immunity of old rats.
