低能量的脉冲染料激光上调成纤维细胞胶原的产生及胶原相关基因mRNA表达

【摘要】 目的观察低能量的波长585nm脉冲染料激光对体外培养成纤维细胞胶原的产生及其某些相关基因的表达的影响,探讨其治疗膨胀纹及美容除皱的可能作用机制。方法以不同能量密度的脉冲染料激光照射体外培养的正常皮肤的成纤维细胞,照射后继续培养24小时及三天,分别以定量PCR(LightCycler)法及化学比色法检测其细胞内不同类型的前胶原基因的表达、TGF-β1及 SMADs mRNA表达与胶原的产生。结果经能量密度为3J/cm2脉冲染料激光照射后的成纤维细胞其上清液中可溶性胶原含量显著高于阴性对照组(p<0.01)及能量密度为4J/cm2组上清液中可溶性胶原含量,在能量密度为3J/cm2时脉冲染料激光能显著促进转化生长因子1、I型前胶原基因α1, α2以及SMAD2,3,4,7mRNA的表达(p<0.05;或p<0.001)。然而,能量密度为4J/cm2的脉冲染料激光对转化生长因子1、I型前胶原基因α1,α2以及SMAD2,3,7mRNA的表达与对照组无明显差异。结论低能量密度(3J/cm2)的脉冲染料激光,能上调成纤维细胞内转化生长因子1及其信号转导分子的SMAD2,3,4,7mRNA的表达,同时增加成纤维细胞内I型胶原基因α1及α2 mRNA的表达, 促进胶原的产生。

【Abstract】 OBJECTIVE: The objective of this study was to evaluate the effect of the pulsed dye laser (585 nm, 450 ms . Cynosure) on the production of collagen and the mRNA expression of collagen relative gene in fibroblasts in vitro. METHODS: Cultured fibroblasts were treated with a 585-nm flashlamp-pumped pulsed dye laser ( fluence 3 J/cm2, 4 J/cm2, respectively, 7 mm spot size ). The production of collagen and the mRNA expression of TGF-β1, SMAD2, 3, 4 ,7 and type I procollagen a1, a2 in fibroblasts were investigated by colorimetry or real time PCR method respectively. RESULTS: The production of collagen was significantly up-regulated after treated with a 585-nm flashlamp-pumped pulsed dye laser in the fluence 3 J/cm2 ( p <0.01) . The mRNA expression of TGF-β1, SMAD2, SMAD3, SMAD4 and procollagen I was significantly up-regulated after treated with a 585-nm flashlamp-pumped pulsed dye laser in the fluence 3 J/cm2 ( p <0.01) . No significantly difference of mRNA expression of SMAD2, SMAD3, SMAD7 and type I procollagea were found between controls and fibroblasts treated with pulsed dye laser in the fluence 4 J/cm2 (p>0.05). CONCLUSIONS: Low fluence (3 J/cm2) pulsed dye laser could upregulated TGF-β1, SMAD2, SMAD3, SMAD4, SMAD7 and Type I procollagen mRNA expression, these might be the reason why it can been effectvely used in the treatment of wrinkles.
