呼吸道合胞病毒G蛋白基因序列测定与分析

【摘要】 目的:掌握浙江地区呼吸道合胞病毒流行株的分子进化及基因变异特点.方法:收集2003-2004年冬季浙江省儿童医院呼吸道感染患儿的鼻咽吸引物,进行病毒分离,对分离株用特异性RT-PCR进行鉴定;对鉴定阳性的RSV毒株进行G蛋白全基因序列测定,并与其他地区RSV分离株的G蛋白基因序列进行同源性比较并构建基因进化树.结果:共分离到4株RSV毒株,3株分离株G蛋白基因全长897bp,另1株为894bp;其中两株的G 蛋白基因序列完全一致;各分离株之间的核苷酸与氨基酸同源性分别为98.05%、97.06%; 与A型RSV毒株标准参考株的核酸同源性为95.69%,与B型RSV参考株的同源性为78.79%;其3’端基因高变区核苷酸序列与GA2亚型的同源性最高.结论:2003-2004年冬季浙江省RSV流行株属于RSV GA2亚型.

【Abstract】 Objective: To study the molecular epidemiology of respiratory syncytial virus (RSV) in Zhejiang province. Methods: RSV was isolated in the nasopharyngeal aspirates(NPA) collected from children with acute lower respiratory tract infection in the Children’s hospital of Zhejiang province in winter during 2003-2004 and the isolated strains were identified by RT-PCR. The whole region of the G protein gene of the isolates was sequenced and the phylogenetic trees were constructed by comparison these sequences with those in die Genbank database. Resnlts: Total four RSV strains were isolated; The G protein gene of the three isolates was 897bp and the other was 894bp; The sequences of the G protein gene of the two isolates were absolutely accordant. The identity of the G gene and the G protein of the four isolates was 98.05% and 97.06% respectively. The identity of the G gene of the four isolated strains with the standard A type of RSV strain was 95.69% and the identity was 78.79% with the standard B type of RSV strain; The nucleotide sequence of the 3’hypervariable region of the G gene was most identified with the GA2 subtype of RSV. Conclusion: The GA2 subtype of RSV was predominant in Zhejiang province during 2003-2004.
