螺旋藻中性多糖硫酸化及性质研究
Study on the Sulfate and Charater of neutral Polysaccharide from Spirulina Platensis

【摘要】 目的对螺旋藻中的中性多糖进行硫酸化修饰并研究其性质。方法采用氯磺酸-吡啶法对螺旋藻中的中性多糖进行硫酸化修饰;玫瑰酸钠法测定修饰前后多糖中的SO42-含量;利用琼脂糖凝胶电泳、红外光谱法及HPLC 对螺旋藻中性多糖硫酸酯进行性质研究。结果修饰前后的多糖SO42-含量分别为0.17%、20.0%;螺旋藻中性糖硫酸酯的琼脂糖凝胶电泳呈一均一斑点,与其他几种硫酸化多糖相比迁移率有较大差异;外光谱法确定硫酸酯键在1270~1200cm-1 、1000~800cm-1处存在强的特征吸收峰;HPLC 显示出一个主要的单峰。结论此制备方法可行。
【Abstract】 Porpuse Sulfate neutral polysaccharides from Spirulina Platensis and study its charater.Methods A neutral polysaccharide from Spirulina Platensis was sulfated by chlorosulfonicacid-pyfidine method.The content of sulfate group of neutral polysaccharide and polysaccharidesulfate was deteceted by the method of sodium rhodizonate.Using agarose gelelectrophoresis(AGE),FTIR and HPLC to study its charater.Results The content of sulfate groupof neutral polysaccharide and polysaccharide sulfate was 0.17% and 20.0%.The electrophoreticbehaviors showed that in AGE electrophotogram the polysaccharide sulfate had one spot andcompared with other kinds of sulfate polysaecharides,its migration ratio was different.Somestrong characteristic absorption peaks of ester sulphate bonds were observed at1270~1200cm-1and 1000~800cm-1 respectively by FTIR.A major single peak was showed inHPLC.Conclusion The sulfate method was successful.
