应用荧光蛋白磷酸酶抑制法检测水中微囊藻毒素

【摘要】 [目的] 探讨荧光蛋白磷酸酶抑制法在水体微囊藻毒素检测中的应用。[方法] 建立藻毒素(MC-LR)抑制蛋白磷酸酶1活性的标准曲线,由标准抑制曲线得拟合方程y=0.90355/(1+exp(1.71636+1.93314*x)),R2=0.983。可由本试验得出的拟合方程计算样品中可能存在的微囊藻毒素类物质的量。[结果]用该方法测定环境水样中微囊藻毒素类物质的结果与高效液相色谱法相关性较好(r=0.9862,P=0.0003)。[结论]荧光蛋白磷酸酶抑制法具有一定的实用价值,适用于对大批量、低剂量MC水样进行监测。
【Abstract】 [Objective] To study the application of fluorescent protein phosphatase inhibition assay for the Detection of microcystins in water. [Methods] Microcystins are potent inhibitors of protein phosphatase 1, and so microcystins in water can be quantified according to enzyme activity in assay system by using the fitting equation which is established in this study. [Results] The fitting equation: y=0.90355/(1+exp(1.71636+1.93314*x)) ( R2 = 0.983) is worked out according to the standard inhibition curve. And the correlation of detecting results between fluorescent protein phosphatase inhibition assay and HPLC is good (r=0.9862, P=0.0003). [Conclusions] Fluorescent protein phosphatase inhibition assay is a rapid, sensitive detection method, which is good for screening and monitoring a large of water samples containing low concentration of microcystins.
