胚胎期暴露于甲基对硫磷和氯氰菊酯对胎鼠和30天龄子鼠脑细胞DNA损伤影响的研究

【摘要】 [目的] 探讨大鼠出生前暴露于农药甲基对硫磷和氯氰菊酯混配对胎鼠脑细胞DNA的毒性效应。[方法] 48只Wistar大鼠在怀孕第0~16天每日一次采用甲基对硫磷、氯氰菊酯混配农药灌胃,暴露剂量分为对照组、低剂量组、中剂量组、高剂量组4个水平,即为0、0.0230+0.8000mg/kg BW、0.0725+2.5265 mg/kg BW、8.0000+0.2300 mg/kg BW。在怀孕第16天断颈处死孕鼠取出胎鼠脑和出生后30天取出子鼠脑用胰蛋白进行消化,制成单细胞悬液,进行单细胞凝胶电泳,吖啶橙染色分析结果。[结果] 怀孕第16天胎鼠和生后30天子鼠神经细胞DNA损伤程度随着农药剂量水平的升高,DNA损伤程度也加重(F=76.26,P<.0001),特别是在孕16天的胎鼠中、高剂量组这种趋势更加明显,其神经细胞彗星样细胞出现率、DNA损伤值也随着农药浓度的增加而增加:与对照组相比较,16天胎鼠的中、高剂量组和出生后30天的高剂量组脑细胞DNA损伤程度都具有显著性差异(P<.0001):胎鼠脑神经细胞DNA的损伤程度也较出生后30天的严重(F=15.81,P<.0001),在中、高剂量组中胎鼠脑细胞DNA损伤程度较30天更为严重。[结论] 大鼠出生前暴露于甲基对硫磷和氯氰菊酯混配可引起脑细胞DNA链断裂损伤,呈现一定的剂量-效应关系,这种效应可持续很长一段时间。
【Abstract】 [Objective] To explore the DMA damage effects by exposure to combined pesticides of Cypermerthrin and Methylparathrion during brain development in offspring rats. [Method] Wistar pregnant rats were divided into 4 groups, i.e. control group and tree combined pesticides treated groups, which were administrated with vegetable seed oil and combined pesticides of Cypermerthrin and Methylparathrion (0.0230+0.8000mg/kg BW, 0.0725+2.5265 mg/kg BW, 8.0000+0.2300 mg/kg BW respectively from 1st to 15th day of gestation.. Single Cell Gel Electrophos (SCGE or Comet Assay) was utilized to measured DNA damage in offspring rat at the 15th day of gestation and 30 days age respectively. [Result] The DNA damage in neurons in offspring rats was increasing with combined pesticides concentration in offspring rat at the 15th day of gestation and 30 days age(F=76.26, P<.0001), especially at the concentration level of 1/95 LD50, 1/30 LD50. Kg-1.d-1. Cosmetic neuron incidence and arbitrary unit developed with increasing concentration of combined pesticides. Compared with the groups of 30-day-age and the foetus of 16 days, the DNA damage degree of the former is more serious than the latter (F =15.81, P<.0001). [Conclusion] Combined pesticides of Cypermerthrin and Methylparathrion can induce DNA damage during brain development and the damage effect can most probably last for a long time.
