HPV16E7基因疫苗在鼠肌组织中的免疫应答

【摘要】 为观察pCDNA3-HPV16-E7重组质粒经肌肉途径接种后在宿主体内的表达,实验以昆明小白鼠为材料,将实验组鼠经后腿股四头肌注射重组质粒pCDNA3-HPV16-E7,对照组注射pCDNA3,于接种前和接种后的第0、2、4、6周取小鼠股四头肌进行免疫组化染色。结果显示,从第2周开始,在注射部位肌肉的肌膜和肌间隙处可见到棕褐色分泌颗粒,随着时间的延长,实验组小鼠肌肉的肌细胞内棕褐色分泌颗粒变大,数量增多,在肌细胞内颗粒由小范围的聚集到整个细胞内都有分布。对照组则未见阳性颗粒。

【Abstract】 The immunohistochemistry method was used to detect the human papillomavirus (HPV) DNA vaccines (pCD-NA3-HPV16-E7) ,which were injected into the leg muscle of mice with administration of 20 μg per mouse. The pCDNA3-E7 was injected into the leg muscle of mice again in the second and fourth week .The pCDNA3 were injected into the leg muscle of the another group mice every two weeks. The muscle of the mice were adopted before injecting, and in the second, fourth and sixth week. The muscle musters were dyed with the immunohistochemistry method .The results showed that pCDNA3-HPV16-E7 could be expressed. There were some brown grains near the injection sites. The brown grains became more and more much and big from the beginning to the sixth week . The brown grains could be discovered in the slices of the control group.
