【摘要】 目的观察大鼠脑出血后血肿周围神经元线粒体内细胞色素C(Cyt-C)释放规律,及中药复方平肝熄风汤的干预作用。方法用Ⅶ胶原酶脑内立体定位注射诱导大鼠脑出血模型免疫组织化学法检测神经元线粒体内Cyt-C的释放情况。结果大鼠脑出血后血肿周围神经元于6h即有Cyt-C释放,出现胞浆染色的阳性细胞;1d后胞浆染色最浓,细胞形态变得不规则,阳性细胞表达最多;3d后染色变淡,阳性细胞数减少;5d后胞浆染色继续变淡,阳性细胞继续减少,治疗组与模型组比较在1d,3d,5d时阳性细胞减少明显(P<0．01),染色亦较模型组淡。结论脑出血后血肿周围神经元Cyt-C于6h即开始从线粒体释放,1d达高峰后下降;平肝熄风汤能阻制Cyt-C的释放,从而组断细胞凋亡。更多还原

【Abstract】 objective: To observe the effect of Pinggan Xifeng decoction(PGXFD) on the rerease of Cytochrome C(Cyt-C) from neuronal mitochondrial around the hematoma after intracerebral hemorrhage(ICH) in rat and to explore the mechanism of PGXF decoction in the treatment of ICH. Methods: Ⅻ type collagenase was stereotaxically injected into the globas pallidus of rats to build the ICH model. immunohistochemical methods was used to exam the release of Cyt-C from neuronal mitochondria after ICH in rat .Results: Cyt-C was began to release in the neuron around the hematoma after ICH in rat at 6 hours after operation, there were some positive cells that were stained at the cytoplasm around the hematoma; after I day, the cytoplasm was stained strongly, the cell shape becamed abnormal, and the number of positive cells peaked to the top; But at 3 days ,the staining becomed weak, the positive cells decreased; at 5 days ,the staining continue weakened and the positive cells continue decreased, the number of positive cells of the PGXFD group was obviously smaller than that of the model group(p<0.01) and the staining was also weaker than that of the model group. Conclusion: the release of Cyt-C begined at 6 h from neuronal mitochondrial around the hematoma after ICH, it peaking to the top at Id . PGXFD can markedly depress the release of Cyt-C.更多还原
