体内外环境因素对成纤维细胞中丝裂素原激活蛋白激酶的影响

【摘要】 目的:研究体内外环境因素变化对成纤维细胞中信号蛋白表达及细胞生物学特性的影响。方法:用免疫组织化学方法分别检测二组各6例体外培养瘢痕疙瘩来源的成纤维细胞(keloidfibroblast, KFB)和正常皮肤来源的成纤维细胞(natural skin fibroblast, NFB)中p44/42MAPK和Phospho-p44/42MAPK,用Ratio值(Phospho-p44/42MAPK积分光密度值÷p44/42MAPK积分光密度值)评价该信号蛋白的活化程度,进行组间对比;并与组织中成纤维细胞进行比较。结果:KFB中p44/42MAPK的积分光密度值高于NFB(P<0.05),而KFB和NFB之间Phospho-p44/42MAPK的积分光密度值无明显差别(P>0.05),但是趋势上在KFB高于NFB;而p44/42MAPK和Phospho-p44/42MAPK的平均积分光密度值以及Ratio值则是在NFB高于KFB(P<0.05)。已经证实K和NS组织FB中p44/42MAPK的积分光密度值和平均积分光密度值在组间无明显差别(P>0.05),而Phospho-p44/42MAPK的积分光密度值、平均积分光密度值和Ratio值在K明显高于在NS。结论:体外培养的人瘢痕疙瘩和正常皮肤组织成纤维细胞中p44/42MAPK的生物学活性不同:体外环境下,KFB和NFB中p44/42MAPK的生物学活性发生改变。

【Abstract】 AIM:To examine the in-vivo and in-vitro environments effect on the expression and activity of p44/42 mitogen-activated protein kinase(p44/42MAPK, or ERK1/2) in fibroblasts of keloid and natural skin.Methods:In order to analyse the differences, we detect the p44/42MAPK and Phospho-p44/42MAPK in fibroblasts from two groups of each 6 samples of natural skin(NS) and keloid(K) cultivated in cover glass in vitro by immunohistochemistry staining.We comment on the contents, activity intencities and activity levels of p44/42MAPK in fibroblasts accordingly and compare them between two groups.At last, we compare them between the fibroblasts in vitro and in vivo.Results: The integral photo densitometry values of p44/42MAPK are significantly, and Phospho-p44/42MAPK insignificantly higher in keloid fibroblasts(KFBs) than in natural fibroblasts(NFBs).The average integral photo densitometry values of p44/42MAPK, Phospho-p44/42MAPK and the Ratio(integral photo densitometry values of Phospho-p44/42MAPK÷integral photo densitometry values of p44/42MAPK) are significantly higher in NFBs than in KFBs. In the tissues fibroblasts, we have known that the integral photo densitometry values and the average integral photo densitometry values of p44/42MAPK are insignificantly different between keloids and normal skins, but they are significantly higher for Phospho-p44/42MAPK in keloids than in normal skin, and the same for Ratio values.Conclusion: The activities of p44/42MAPK between the KFBs and NFBs in vitro are different, and the same between in-vivo and in-vitro.
