【摘要】 目的:采用高效毛细管电泳法(Capillary Electrophoresis,CE)检测海洛因依赖者尿吗啡的含量.方法:BECKMAN毛细管,内径50 μm,长度67cm;缓冲液:0.05mol/L磷酸氢二销-磷酸二氢钠:乙腈(80:20),pH=6.9:缓冲液温度:20℃:紫外检测波长:214nm:衡压分离,分离电压:15.00kV;自动气压进样,20pis,5sec.结果:在吗啡浓度为2μg/ml-1000μg/ml 范围内,线性良好,回归方程为Y=33.2928X+125.3384,r=0.999,Y为峰面积,X为浓度(μg/ml).吗啡标准检测的日内相对标准偏差为2.58%,日间相对标准偏差为4.41%;加样回收率为90.46%,RDS为7.18%.采用此方法检测了5例海洛因依赖者脱毒前后的尿吗啡含量,脱毒前分别为34.35μg/ml、46.82μg/ml、60.81μ/ml、25.72μg/ml和44.58μg/ml,脱毒后期(末次吸毒后150小时)分别减少为0.44μg/ml、0.52μg/ml、1.32 μg/ml、0.15μg/ml和0.52 μg/ml.结论:此方法重现性好,灵敏度高,对海洛因依赖者的尿吗啡可进行定性、定量检测,可作为海洛因依赖者诊断和脱毒疗效评价的有效手段.更多还原

【Abstract】 Objective: To establish a quantitative determination method for morphine concentration in heroin addicts urine by CE. [Methods]: CE consisted of BECKMAN capillary ,50 μ m x 67 cm , automated sampling and UV-detector; running buffer included 0.05 mol/L Na2HPO4-NaH2PO4 buffer : acetonitrile (80:20 v/v),pH 6.9 and the running buffer temperature was set at 20℃. The detector wave-length was set at 214nm . Injection pressure was 20 pis, 5 sec and power supply was 15kV constant. [Results]: The standard curve was linear in the morphine concentration ranging from 2 μ g/ml to 1000 μ g/ml, regression formula was Y=33.2928 X + 125.3384, Y represents peak area , X represents morphine concentration (μ g/ml) , and correlation coefficient was 0.999 . The average recovery rate of morphine was 90.46%, RDS was 7.18%. Five cases of morphine in heroin addict urine were determined by the method , the morphine concentration were 34.35 μg/ml ,46.82 μ g/ml , 60.81 μ g/ml , 25.72 μ g/ml and 44.58 μ g/ml respectively at 3.5 ,2.5 , 4.5 , 8.0 and 5.0 hours after last taking heroin and the morphine concentration were decreased to 0.44 μ g/ml ,0.52 μg/ml ,1.32μ g/ml ,0.15 μ g/ml ,0.52 μ g/ml at 150 hours after detoxification . [Conclusion]: The results showed that the method was excellent reproducibility and high sensitivity , which could be used to diagnose the morphine dependence and evaluate the detoxification efficiency.更多还原
