RP-HPLC法测定苦皮藤愈伤组织中苦皮素A的含量

【摘要】 目的:建立了利用RP-HPLC法测定愈伤组织中苦皮素A的含量。方法:采用Shim-pack CLC-ODSC18色谱柱,甲醇—水(90:10)为流动相,流速为1ml/min,检测波长为254nm,对苦皮藤愈伤组织中苦皮素A的含量进行测定。结果:苦皮素A标准品在0.06~0.74ug范围内与峰面积呈良好的线性关系;精密度试验相对偏差为0.87%和1.41%;平均加样回收率为96.21%;测得了所取样品苦皮藤愈伤组织中苦皮素A的含量为0.12%(鲜重)。结论:该方法操作简便,易行,线性关系良好。
【Abstract】 AIM: The purpose is to develop a method the contents of Angulatin A in the callus culture of Celastrus Angulatus M by RP-HPLC. METHOD: The method was used Shim-pack CLC-ODS C18 column with MeOH-H2O(90: 10) as the mobile phase, 1ml/min as flow-rate and the photodiode array detector at 254 nm. RESULTS: Calibration graphs was rectilinear between 0. 062ug and 0. 74ug under the experimental conditions. The recovery was 96. 21%. The relative standards deriations (RSDs) of measurement precision tests were 0. 87% and 1. 41%. The content of Angulatin A was 0. 12% (Fresh Weight). CONCLUSION:The method was simple and had good liner relationship.
