High-throughput foetal DNA separation from maternal plasma by hybridisation capture and magnetic particles

【摘要】 <正>Objective To develop a high-throughput method for foetal DNA separation from maternal plasma which allows enrichment of small (mainly foetal) DNA fragments. Meh-tod A robot was programmed to automatically separate specific DNA fragments after hybridisation capture and attachment to bio-magnetic particles. Plasma pools from >30 D-negative pregnant women at 28 ~ 30 week of gestation send for antibody screening were used. Real time PCR for RHD and RHCE exon 7 was applied. Statistics of at least 8 replicates were performed using SPSS software. The a-mount of foetal and maternal DNA in maternal plasma was calculated from a curve using human DNA standard.
