纯度对抑制素pcISI基因免疫黄牛的免疫反应性和黄体发育的影响

【摘要】 目的:探讨两种不同纯度的双拷贝抑制素真核融合表达质粒peISI(由抑制素α1-32基因与乙肝表面抗原(HbsAg)基因构建)免疫对黄牛的抗体P/N值、抗体阳性率和黄体发育的影响。方法:将四种不同剂量(0.75mg、1.5mg、2.25mg、3.0mg)的高纯度抑制素融合质粒pcISI和四种不同剂量(0.75mg、1.5mg、2.25mg、3.0mg)的低纯度的抑制素融合质粒pcISI免疫南阳黄牛。21天后加强免疫。结果:高纯度pcISI(OD260/OD280介于1.8-2.0)的免疫效果略高于低纯度组(OD260/OD280介于1.7-1.8和2.0-2.1)(P>0.05)。高纯度质粒组黄体略大于低纯度组(P>0.05)。2.25mg抑制素融合表达质粒组的抗体P/N值和抗体阳性率明显高于0.75mg组(P<0.05)。结论:质粒纯度不能明显影响双拷贝抑制素α1-32基因与乙肝表面抗原(HbsAg)基因构建的抑制素真核融合表达质粒pCISI免疫黄牛的免疫反应性和黄体发育能力。
【Abstract】 Aim:To discuss effect of two different purity of inhibin gene expressing plasmid pcISI on immunoreactivity and corpus iuteum of the scalper.Method:The scalper were immunized with the different dosage (0.75mg,1.5mg,2,25mg and 3.0mg) of highly-or lowly-purified inhibin fusion expressing plasmid pcISI.Result:In either PIN value or rate of the positive antibody against inhibin,the group with highly-purified plasmid (OD26./OD280 between 1.8 and 2.0) was a little higher than that with the lowly-purified one(OD260/OD280 between 1.7 and 1.8 or between 2.0 and 2.1)(P>0.05),the same as the corpus luteum development.Whatever P/N value or rate of the antibody against and 0.75mg one was inhibin,dosage of 2.25 mg was the best of all in the experiment.The difference between 2.25mg dosage-group very vivid (P<0.05).Size of the corpus luteum of the yellow cattle was monitored by B ultrasonic diagnosis during the early embryo development stage.The results illustrate that:purity didn’t affect either imunoreactivity or corpus luteum’s development.
