海拔压力与中国牛属MSTN基因变异

Altitude Pressure on the Genetic Variation of Myostatin Gene in Bos Species in China

【摘要】 针对牛肌肉生长抑制素基因(MSTN)基因三个外显子区设计引物,通过PCR扩增及测序,以蒙古牛、雷琼牛、独龙牛、巴音郭楞州牦牛共66个样本为材料检测了中国普通牛、瘤牛、大额牛和牦牛4个家牛属物种该基因外显子区核苷酸序列的变异。结果显示,4个中国牛种的MSTN基因三个外显子分别含375,372,381个碱基对,在所检样本中,发现8个核苷酸多型位点,定义15种单倍型;种内核苷酸多样性(Pi)在0.00036-0.00103间,种间核苷酸多样性(Dxy)在0.00121-0.00499间。另外,Dxy值与各群体中心产区的海拔高度相关性显著,表明海拔对MSTN基因的分化具有一定的压力作用。

【Abstract】 Using the methods of PCR amplification and sequencing,variations of the myostatin(MSTN) gene were detected in 4 Chinese Bos species,normal cattle,zebu,yak,and gayal.The results showed that,375,372,381 base pairs were detected respectively in the three exons of the MSTN gene in the 4 Bos populations;8 nucleotide polymorphic sites were observed with 15 haplotypes defined,and the nucleotide diversities within the 4 species(Pi) ranged from 0.00036 to 0.00103,and that between the 4 species(Dxy)ranged from 0.00121 to 0.00499;furthermore,the Dxy values correlated significantly with the altitudes of the central habitats for each population.The correlation between altitude and the nucleotide diversities indicated that altitude exerted a certain pressure on the genetic variations of the MSTN gene in Bos species.
