【摘要】 本实验的主要目的是检测重组 T4噬菌体经肌肉注射以后,在鸭体组织器官的分布和存留情况. 首先,用小量的液体培养基培养重组噬菌体 T4-VP2,再通过扩大培养、浓缩后获得滴度为2.81×10¹⁰ 个/ml 的重组噬菌体 T4-VP2。然后,用重组噬菌体 T4-VP2进行动物实验。将12只5d 龄的小鸭分为2组:A 组6只,为肌肉注射组,每只小鸭注射250uL 的重组噬菌体;B 组6只,为空白对照组, 不作任何处理.分别在处理后第1、2、3h 及第2-10d 取小鸭粪便,在处理后的第10h 和1、3、5、7、 9d 剖杀小鸭,取出内脏,进行噬菌体检测,并对分离的噬菌体进行 PCR 鉴定.实验结果表明重组 T4 噬菌体能很好地进入小鸭的血液循环系统,在鸭的心、肝、脾、肺、肾等组织器官和小肠内容物、血清中都有存留,而且它在这些部位的存留时间各不相同,最短的有3d,最长的有7d.这些结果初步反映出其作为新型基因工程活疫苗的可能性,为研制高效、廉价的活疫苗提供有效数据和实验基础.更多还原

【Abstract】 Through the animals’ experiment,we can fred out that fate of recombinant T4 phage after injection into the muscle of the ducks.First,through using small liquid culture to make phage,amplifying and concentration,we can amplify a lot of recombinant phages,which had 2.81×10¹⁰ phage/ml.Second,in this animal experiment,we used recombinant phage T4-VP2 as materials.Twelve five-day-old ducks were randomly divided into two groups.Group A had 6 ducks and received feeding of 250uL T4-VP2 each. Group B had 6 ducks and served as control,the ducks in which received nothing,only for comparison. After dealing with different methods and PCR’s detection,we can found out that the recombinant phage T4-VP2 can entered into the ducks’ blOod system,survived in the ducks’ heart,liver,lung,kidney and so on. It can also survive in the ducks for a long time.It was reflected that the possibility of live recombinant vaccine by using phage T4.更多还原
