猪SIM1基因部分序列测定及定位研究
SM1 Gene Sequencing and Mapping Using Radiation Hybrid Panel in Swine

【摘要】 SIM1(single-minded1)基因在中枢神经系统的形成及中线细胞发育中具有重要作用,它的突变可导致下丘脑视上核和室旁核神经内分泌细胞功能失调,影响动物的采食行为, 从而导致肥胖和体重增加。SIM1基因已经在人和鼠上定位。本研究根据人SIM1基因外显子区段(exon7-exon8)设计引物,对猪基因组进行PCR扩增、测序。结果表明:扩增的序列长710bp, 包括部分exon7、部分exon8和完整的intron7。与人、鼠相应序列比较,同源性分别为62.25 %和54.69%。采用IMpRH panel将SIM1基因定位在猪1号染色体1p13上。
【Abstract】 SIM1 (single-minded 1) gene plays a key role in the formation of central nervous system and the midline cell linege. Mutation of SIM1 failed supraoptic and paraventricular nuclei of the hypothalamus capability, therefore affected animal feeding behavior, obesity and fat deposition. SIM1 has been mapped on human and mouse. In present study, primers amplifying porcine SIM1 gene were designed based on exon7 to exon8 sequences of the published human SIM1 gene. The PCR product was sequenced. The sequence length is 710bp, including partial exon7, partial exon8 and complete intron7. Compared with that of human and mouse, the rates of identity are 62.25% and 54.69%. Using IMpRH. panel SIM1 gene was mapped to chromosome 1p13 in swine.
