【摘要】 选取生产性能具有明显差异的4个猪品种(大花白、二花脸、杜洛克和长白)构建品种DNA池,采用测序的方法研究猪CTSD基因3’非翻译区(426bp)序列的多态性,快速筛查到2个SNF位点(A-1664G和C-1930A)。进一步对A-1664G位点利用MlsI内切酶进行 PCR-RFLP和利用测序图中SNF等位基因峰高的比值估算各猪品种等位基因的频率,结果表明猪CTSD基因3’LITR区MlsI位点中3种基因型在4个品种中分布在中外猪种中卡方检验差异显著(P<0．05):(1)AA基因型频率在外国猪种(杜洛克和长白)中明显高于中国猪种(大花白和二花脸);(2)A等位基因频率在外国猪种中分布接近1．0,而在中国猪种中接近0．5,显示该位点可能是一个用于分析中外猪种性能差异的分子标记;(3)在4个品种中,大花白、二花脸、杜洛克和长白猪中不同基因型的分布均符合Hardy-Weinberg平衡(P>0．05)(4)卡方检验用2种方法得到的频数X2值,差异均不显著(P<0．05),说明测序峰高比值估算的等位基因频率与RFLP得到的基因频率基本相符。更多还原

【Abstract】 Four breeds of pig (Dahuabai,Erhualian, Duroc, Landrace ) with different production were applied to screen potential single nucleotide polymorphisms (SNPs) based on DNA pooling and sequencing. Two SNPs A-1664G and C-1930A. were found successfully in the fragment of 426 bp at the 3’ untranslated region.. The genotype of A-1664G MlsI site were tested by PCR-RFLP and calculating the ratio of allele peak heights in sequencing draft.The result showed: (1)The genotype frequency of AA in foreign breeds (Duroc and Landrace) was significantly higher(P<0.05) than that of the native breeds.(Dahuabai,Erhualian);(2)The allelic frequency of A was approximately 1.0 in foreign breed ,but 0.5 in native breed.This result demonstrated that MlsI site may be a potential molecular marker for different production;(3)Genotype frequencies in four breeds were all observed Hardy-Weinberg principle((P<0.05);(4) The accuracies of estimation for the allelic frequencies of A and G were verified by compariong with the results of PCR-RFLP and no significant different was found between two methods based on X2 test.更多还原
