五指山猪等5个猪品种IGF2基因部分序列的SNP分析

【摘要】 利用PCR产物直接测序法,对五指山猪、滇南小耳猪、香猪、大白猪和梅山猪共 60个样本的胰岛素样生长因子2基因5’调控区部分片段的单核苷酸多态性进行了研究。结果找到10个SNP,分别是:C5872T、C5888T、A5976G、C6010T、T6029A、C6037T、C6043T、 C6063T、C6112T和C6164T。所有位点均为二等位基因型,T6029A为T←→A碱基颠换,A5976G 为A←→G转换,其他均为C←→发转换。统计各位点等位基因和等位基因型在总群体与各品种内的分布频率,发现在A5976G位点,五指山猪、滇南小耳猪和香猪的优势等位基因均是G, 而大白猪和梅山猪该位点优势等位基因均为A;在C6164T位点,三个小型猪的优势等位基因均为T,而另两个猪品种该位点优势等位基因都是C;A5976G处于IGF2基因外显子1区域, 可能直接或通过改变启动子1的组成而引起IGF2结构和功能发生变化。并针对这10个SNP 位点进行组合基因型分析,找到21种组合基因型,发现Hap 18型(TCGCTTCTTT型)仅出现于三种小型猪中,而Hap 14型(TCACTCCTTC型)仅出现于大白猪和梅山猪中。结果反映, A5976G与C6164T SNP位点和Hap 18型与Hap 14型组合基因型可能与小型猪的矮小机制有关。
【Abstract】 Single nucleotide polymorphism of partial 5’ UTR of IGF2 gene in 60 pigs from Wuzhishan, Diannan, Xiang, Large White and Meishan pig breed was studied by the sequencing method. As a result, 10 SNP sites were detected, which were C5872T, C5888T, A5976G, C6010T, T6029A, C6037T, C6043T, C6063T, C6112T and C6164T. Every site was biallelic, only 1 transversion was observed at T6029A site, with 9 transitions at other sites, including 1 A←→G transition and 8 C←→T transitions. The gene and genotype frequencies of every SNP sites in the whole group and in each breed were counted respectively, the results showed that the predominant allele was G at A5976G site in Wuzhishan pig, Banna pig and Xiang pig, but it was A in Large White pig and Meishan pig; The predominant allele was T at C6164 site in 3 miniature pigs, however, it was C in other 2 breeds; Besides, A5976G was located in exonl, which perhaps influenced the function of IGF2 through influencing the structure of the promoter 1. We also performed the composite genotype anlysis on the 10 SNP sites, and 21 kinds of composite genotypes were observed, in addition, Hap 18 type(TCGCTTCTTT) was only found in all 3 miniature pigs, on the contrary, Hapl4 type(TCACTCCTTC) was only found in Large White and Meishan breeds. All the results revealed that the SNP sites A5976G and C6164T, and the composite genotype Hap 18 and Hap 14 were possibly related to the drawfism mechanism of the miniature pigs.
