五龙鹅良种选育中分子标记的研究
Study of the available genetic marker in breed Wulong geese

【摘要】 以著名的地方特色品种五龙鹅和隆昌鹅组合杂交、筛选出优化的杂交后代,用 (SDS)PAGE对五龙鹅及杂交后代的血液蛋白质进行多态分析。用改进的聚丙烯酰胺凝胶系统对五龙鹅及杂交后代种的血液碱性磷酸酯酶(AKP)以及血液淀粉酶(Amy)进行遗传变异多态性检测,并对其进行了表型、基因型和基因频率的统计和计算。结果表明,在还原条件下,全血样品更能显示出蛋白质的多态变化,优化杂交后代鹅的蛋白质结构出现新的亚基组分S1-S3,清蛋白(Alb)和转铁蛋白(Tf)受多等位基因支配,其差异变化的频率直接或间接与生产性能相关。在AKP位点检测到快、慢三条带,为2个共显性遗传的等位基因Akp和asp控制,五龙鹅生产性状特征与表型带谱频率具有相关性。在Amy-1位点检测到AA、AB和BB 3种类型,为4 个共显性遗传的等位基因Amy-1A、Amy-1B、Amy-1C和Amy-1D控制,并分析了五龙鹅豁眼性状特征与表型带谱频率的相关性。
【Abstract】 The crossing combinations were conducted by the famous local feature breed geese, Wulong geese and LongChang geese, and then screened dominance hybrid progenies.The blood protein polymorphisms of the hybrid progenies were determined via (SDS)PAGE. Alkline phosphatase was detected AKP areas (or locus) via polyacrylamide gel elctrophoresis(PAGE). The polymorphisms and genetic variation of amylase were investigated Via polyacrylamide gel elctrophoresis(PAGE). The result showed that in reduced condition whole blood samples appeared more protein polymorphism than red cell or serum. In the constitutional proteins of dominance hybrid female progenies the new subunits such as S1-S5 showed, and exhibited higher polymorphism. On the other hand, the albumin and transferrin locus (Alb/ Tf) were controlled by multi-alleles. The frequency of the difference changes could directly or indirectly regulate and affect on the productivity of the hybrid progenies. The frequency of phenotype and genetype and gene for AKP areas were counted in this paper. AKP was scored F and S types and controlled by 2 co-dominant alleles (AKP, akp), and the relativity of huo yan traits of productivity to zymogram(band) frequency in the geese was discussed in this paper. The frequency of phenotype, genetype and gene for Amy-1 areas were counted. Amy-1 was scored AA, AB and BB types and controlled by 4 co-dominant alleles (Amy-1A, Amy-1B, Amy-1Cand Amy-1D), and the relativity of huo yan traits to zymogram(band) frequency in the geese discussed.
