北京鸭与樱桃谷鸭遗传多样性的微卫星分析

【摘要】 用双重抑制PCR技术分离7对鸭的特异微卫星引物,,对北京鸭A、B、C三个系和樱桃谷鸭进行了遗传多样性分析,结果表明:所用的7对引物中4对呈高度多态,3 对呈中度多态。群体平均杂合度最高的是北京鸭A系,0.5780;杂合度最低的是樱桃谷鸭, 0.5616,在DA遗传距离上,北京鸭B系与樱桃谷鸭遗传距离最近:0.0244,北京鸭A系与北京鸭C系遗传距离较远:0.0946。在UPGMA聚类图上北京鸭B系先和樱桃谷鸭聚在一起,然后和A系聚在一起,最后再和C系聚在一起。说明北京鸭经过不同方向选育后,C 系和A、B系之间出现了差异。本研究结果与育种实际相符合,也从另一方面说明了品系的遗传特征能够由各微卫星位点的多态性得到真实的反映。
【Abstract】 Dual-suppression-PCR technique was used to isolate seven special duck microsatellite primers, by which the Genetic Diversity of Cherry Valley duck and three strains (A,B,C) of Beijing duck were analyzed. The results indicated that there were four loci with high polymorphism, three loci with middle polymorphism. The highest population mean heterozygosities was A strain of Beijing duck, which was 0.5780;and the lowest was Cherry Valley duck, which was 0.5616.As for the DA genetic distance, B strain of Beijing duck was closest to Cherry Valley duck, which was 0.0244, and A strain was the minor, which was 0.0946. B strain of Beijing duck was first grouped with Cherry Valley duck, then with A strain, lastly with C strain in the UPGMA/ DA dendrogram, which indicated that after different direction selection of Beijing duck, there has been difference among the strains of A,B,C. The results of the study conforms the breeding practice, which indicated that the genetic characteristics could be reflected by the microsatellite polymorphism in another respect.
