【Abstract】 The culture methodology of primary mammary epithelial cells in Holstein Cows has been studied in this paper. The primary mammary epithelial cell derived from biopsy tissue obtained from a lactating-period Holstein cow. The tissue was minced and digested by trypsin-collagenase. The mammary epithelial cells were collected by centrifuge. It was showed that the primary mammary epithelial cell can be isolated from the tissue and purified by trypsin digestion finely. The cells tend to form monolayer aggregates with the characteristic cobblestone of epithelial cells. Immunocytochemistry with keratin antibodies was positive. When passaged, the mammary epithelial cells grow well and proliferate vigorously in this culture system.更多还原
