不同硼水平对固始鸡血细胞的影响

【摘要】 目的:研究不同硼水平对固始鸡红、白细胞数,各种白细胞比例及血细胞形态结构的影响。方法:320 羽1d固始鸡,按体重相近原则随机分为4组,在饮水中添加0,100,200,400 mg·L-1硼,试验期6w。每周末每组随机选取固始鸡10羽,颈静脉采血,红细胞计数、白细胞计数,血涂片制作,瑞氏-吉姆萨氏染色, 白细胞分类计数,油镜观察和显微摄影。结果:(1)各试验组固始鸡红、白细胞数明显低于同日龄对照组; 饮水中添加100mg·L-1硼对固始鸡各种白细胞比例影响多不显著,高硼(硼添加量≥200 mg·L-1)对嗜碱性粒细胞、淋巴细胞和单核细胞比例影响明显。(2)各种血细胞形态结构变化明显,红细胞大小不一,形态各异, 细胞核固缩、肿胀、断裂或解体,白细胞体积明显变小,细胞核固缩、溶解、消失,其中有粒白细胞胞质颗粒明显减少或解体。结论:结果表明不同硼水平,尤其高剂量硼,可明显降低固始鸡的红、白细胞数及嗜碱性粒细胞和单核细胞比例,对各种血细胞形态结构有明显影响,甚至出现损伤作用。

【Abstract】 Objective: To study the Effects of different level of Boron on the RBC and WBC numbers, the WBC percentage and the hemocytes shape and structure in Gu-shi chickens. Methods: A total of 320 one day-old Gu-shi chickens were randomly divided into four groups, and watered supplemented with 0, 100, 200, 400 mg·L-1 boron for six weeks. Ten chickens in each group were selected randomly and the blood were got by jugular vein at each weekend; the blood films were made and stained with Wright-Giemsa mixed staining solution; the red cells count(RCC), white cells count(WCC) and the hemomyelogram were carried out; the shape and structure of hemocytes were observed and shoot in Gu-shi chickens. Result: (1) Compared with control group of the same age, the RBC and WBC numbers decreased obviously, the effects of boron on the percent of WBC in experiment groups were not significant (p≥0.05) adding 100 mg·L-1 boron in drinking water, but there were obvious effects on the percent of basophile granulocyte, lymphocytes and monocyte with the high dosage Boron(≥200 mg·L-1). (2)The changes of erythrocytes and leucocytes shape and structure were obvious, the RBC size and shape were diverse and the nucleus of them swelled, shrinked or disorganized; the WBC minished obviously in volume and the nucleus of them shrinked, the granules number in granulocyte decreased obviously or even disappeared. Conclusion: The results showed that the different level boron, especially high dose boron may decreased obviously the number of RBC and WBC numbers and the percent of basophile granulocyte and monocyte, affected obviously and even damaged severely the shape and structure of all kinds of blood cells.
