小鹅瘟GPV-CHa15弱毒株培育及其特性研究

【摘要】 以小鹅瘟病毒(GPV)CHv强毒株在10日龄鸭胚上传代获得的GPV-CHa15弱毒株的ELD50为106.84/0.3ml(10日龄鸭胚),对1日龄雏鹅没有致病性;分别通过雏鹅和成年鹅体各10代无返强现象;接种雏鹅后能够在雏鹅体内繁殖并向环境排毒,使同居雏鹅感染并获得一定程度免疫力;对雏鹅和种鹅的最小免疫剂量为1000倍ELD50/0.3ml;雏鹅接种后第3天即有95%(19/20)能够抵抗100个LD50/0.3ml(10日龄雏鹅)强毒攻击,第5天后即能100%获得保护;母鹅免疫注射后第15天的后代雏鹅即可100%抵抗强毒攻击;雏鹅免疫途径以皮下和肌肉注射途效果最好,口服效果次之,滴鼻和眼滴效果较差;成年种鹅免疫途径以皮下和肌肉注射效果最好,滴鼻和眼滴效果次之,口服效果较差;对GPV-CHv强毒株在雏鹅体内的繁殖和致病有干扰作用;对雏鹅的免疫期在30天以上;对成年种鹅的免疫期可达5个月;第45和42代分别对雏鹅和种鹅的免疫原性开始减弱。经临床使用结果进一步表明GPV-CHa15弱毒株安全、稳定性好、免疫原性良好。

【Abstract】 The results of studying on the growth of attenuated vaccine strain( GPV-CHa15 )of Gosling Plague (GP) and its characteristic are report in this paper. An attenuated vaccine strain (GPV-CHa15) was obtained by passage in 10-day duck embryo for 15 passages. The strain’s characteristic as fellow: ELD50 is 106.84/0.3ml (10-days duck embryo), invimlence to one day old gosling, and not became virulence when growth in goslings and geese for 10 passages. The strain can reproduce in gosling’s body after inoculated, and excreted into environment and infected goslings can get some immunity. Minimum immune dosage to goslings and geese is 1000 ELD50/ 0.3m l ( 10-daysduckembryo) . 95 percent( 19/20) goslings can against 100LD30/0.3ml ( 10-days goslings) virulence strain (GPV-CHv )3 days post inoculated( PI) and 100 percent(20/20) goslings can against 100LD50 / 0.3ml (10-days goslings) GPV-CHv strain 5 days PI. 100 percent(20/20) goslings produced from parents that 15 days PI can against 100LD30/0.3ml ( 10-days goslings) . The best immune effects to gosling are hypophloeodal injection and muscle injection, and orally, nose dropping and eyes dropping are next. The best immune effects to geese are hypophloeodal injection and muscle injection, and nose dropping, eyes dropping and orally are next. Reproduce and virulence could be affected of GPV-CHv virulence strain by GPV-CHal 5 strain when infect goslings in the same times. Immunity time could be last over 30-days to gosling and about 5 months to geese. Immunity become lower to gosling when GPV-CHal 5 strain pass 10-days duck embryo over 45 passages and to geese vvhen GPV-CHal5 strain pass 10-days duck embryo over 42 passages. GPV-SCal5 is stabilization, safety with excellent immunity.
