饲粮铜源和水平对断奶仔猪垂体生长激素mRNA水平的影响
EFFECTS OF DIETARY COPPER SOURCE AND LEVEL ON PITUITARY GROWTH HORMONE MRNA LEVELS IN WEANLING PIGS

【摘要】 用平均35日龄断奶、体重9.5千克的长白×大白杂交阉公猪30头进行试验,研究饲粮铜源和水平对断奶仔猪垂体生长激素mRNA水平的影响。试猪按完全随机区组设计依体重和窝别随机分为3个处理组,分别饲喂添加15和250mg/kg铜(硫酸铜)及250(mg/kg)铜(甘氨酸铜)的饲粮。猪只个体饲养于代谢室(22.5℃左右)内无元素污染的玻璃钢代谢笼中,试验期28天。铜源和铜水平对猪生长速率、采食量、饲料转化效率、血红蛋白浓度、血细胞压积、血浆生长激素水平、血浆生化指标(葡萄糖、白蛋白、球蛋白、总胆固醇、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇、甘油三酯和碱性磷酸酶)及血浆、胰、肾、脾中铜含量均无明显影响(P>0.10)。但是,与添加15和250mg/kg硫酸铜来源的铜组相比,添加250 mg/kg甘氨酸铜来源的铜显著提高了断奶仔猪垂体生长激素mRNA水平(P<0.007)。与添加15mg/kg硫酸铜来源的铜组相比,添加250mg/kg硫酸铜和甘氨酸铜来源的铜均明显提高了断奶仔猪脑、肝及心肌中的铜含量(P<0.002),而两种铜源间无明显差异(P>0.60)。本次研究结果表明,铜能通过促进垂体生长激素的基因表达而促进幼猪的生长;现代分于生物学指标-垂体生长激素mRNA水平在反映机体生长激素确切状况上比传统的常规血清或血浆生长激素水平更敏感;甘氨酸铜在提高断奶仔猪垂体生长激素mRNA水平上比硫酸铜更有效。
【Abstract】 An experiment was conducted using a total of 30 crossbred (Landrace x Large White) weanling barrows averaging 9.5 kg BW and 35 d to investigate the effects of dietary Cu source and level on pituitary growth hormone (GH) mRNA levels. Pigs were randomly allotted by bodyweight and litter to one of three treatments in a randomized complete block design, and fed basal corn-soybean meal-whey diets supplemented with 15 and 250 mg/kg Cu as CuSO4· 5H2O or 250 mg/kg Cu as Cu-glycine, respectively. Pigs were individually kept in glass fiber metabolism crates in a metabolism room (around 22.5 ℃) for the duration of the 28-d experiment. Either Cu source or Cu level had no effect (P>0.10) on growth rate, feed intake, feed conversion efficiency, hemoglobin, hematocrit, plasma GH level, plasma biochemical indices (glucose, albumin, globulin, total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, and alkaline phosphatase), and Cu contents in plasma, pancreas, kidney and spleen. However, the addition of 250 mg/kg Cu as Cu-glycine to the diet increased (P<0.007) pituitary GH mRNA level of weanling pigs compared to the addition of 15 and 250 mg/kg Cu as CuSO4 · 5H2O. The addition of 250 mg/kg Cu as either CuSO4 · 5H2O or Cu-glycine increased (P<0.002) Cu contents in brain, liver and heart compared to the addition of 15 mg/kg Cu as CuSO4 · 5H2O with no difference (P>0.60) between the two Cu sources. The results from this study indicate that Cu could enhance the growth of young pigs by promoting the pituitary GH gene expression; The modern molecular biology’s index-pituitary GH mRNA level was more sensitive than traditional conventional serum or plasma GH level in reflecting the exact GH status in the body; And organic Cu-glycine was more effective than inorganic CuSO4 · 5H2O in increasing pituitary GH mRNA level in weanling pigs.
