乳酸杆菌表达的重组降钙素生物学活性研究

【摘要】 用SD系大白鼠对乳酸杆菌表达的基因工程降钙素的生物学效应进行了研究.第一次预实验结果显示.重组乳酸杆菌LaS318培养上清可使SD大白鼠血钙由10.5mg/100mL下降为9.4mg/100mL证明LaS318培养上清中的降钙素具有降低大白鼠血钙的作用.第二次预实验结果显示,LaS318培养上清中降钙素的生物学效价接近4mU/mL。然后,利用降钙素国际标准品,通过平行线原理四点法(2.2法)测定了LaS318培养上清中降钙素的生物学效价为2.72mU/mL,其95%可信效价范围为2.18-3.24(mU/mL),平均可信限为19.42%。
【Abstract】 Biological activity of the recombinant Calcitonin (rCT) was determined by Rats Hypercalcemic Assay. The first experiment showed that rat hemocalcium decreased from 10. 6mg/100mL to 9. 4mg/100mL post injecting culture supernatant of the recombinant Lactobacillus. The second experiment showed that the biological activity of rCT was close to 4 mU/mL. At last, the biological activity of the rCT in culture supernatant of the recombinant Lactobacillus was determined to 2. 72mU/mL by Parallel-Principle-Four -Points-Method (2. 2 Method ), using WHO IRP of CALCITONIN, SALMON. The 95% FL of PT was 2.18 - 3. 24mU/mL, the FL% was 19. 42%, The data suggested that the result was accurate, the error was on the small side.
