新城疫病毒鹅源毒株ZJ1株全基因组的序列测定

【摘要】 依据GenBank上公布的鸡新城疫病毒(NDV)的基因序列,自行设计了9对引物。运用反转录-聚合酶链反应(RT-PCR)获取覆盖NDV鹅源毒株ZJ1株全基因组的重叠片段。病毒RNA的5’及3’端的序列由cDNA末端快速扩增技术(RACE)获取。整个病毒基因组序列由15192个碱基组成,比GenBank上公布的其他4个NDV毒株Beaudette C、B1、La Sota及Clone-30的全基因序列长6个核苷酸,这6个核苷酸位于核衣壳蛋白(NP)基因内,相对于NDV毒株La Sota株全基因序列的1647nt-1648nt位,其他9个NDV毒株也被验证是否存在此6个额外的核苷酸。实验结果表明共8个NDV毒株具有此特征.他们分别属于3个不同的基因型。
【Abstract】 On the basis of comparing the sequences of Newcastle disease viruses (NDV) genes released in GenBank, 9 pairs of primers were designed and utilized to acquire overlapping viral cDNA fragments of a NDV of goose origin (ZJ1) by high -fidelity reverse transcription-polymerase chain reaction (RT -PCR). The sequences of the 3’ - and 5’ - terminal ends of viral RNA genome were determined by sequencing cDNA fragments generated by rapid amplification of cDNA ends. The entire genomic sequence consisted of 15192 nt, which was 6 nt longer than the published full length NDV genome of strains La Sota, B1, Clone -30 and Beaudette C. This additional 6 nt motif was located in the non-coding region of nucleoprotein (NP) gene be-tween the corresponding positions 1647nt and 1648nt on the NDV genome of strain La Sota. Other 9 NDV strains were also searched for the existence of this additional 6 nt motif. The results showed that 8 NDV strains of 3 different genetic groups shared this characteristic.
