鸡毒支原体油乳剂灭活疫苗的研制

【摘要】 通过高速离心或中空纤维滤器浓缩鸡毒支原体CR株培养物的方法浓缩抗原,灭活后加入矿物油佐剂,经乳化制成疫苗9批,然后分别将这些疫苗免疫接种鸡,测定其安全性和免疫效力。试验结果表明:1.两种抗原浓缩方法制成的疫苗均安全有效,差异不大;2.用5倍浓缩的抗原制成疫苗便能使鸡产生良好的免疫力,免疫保护力随着抗原浓度提高而提高。而用不浓缩的原倍培养物制成的疫苗几乎不能使鸡产生免疫保护力。以两倍免疫剂量接种,所有各批疫苗对鸡均不产生副反应,疫苗对鸡安全。疫苗接种鸡体内产生的抗体能维持到10个月以上,免疫鸡在强毒攻击后气管中支原体分离率明显低于对照鸡。

【Abstract】 Both methods of centrifugation and filtration were used to concentrate Mycoplas-ma gallisepticumCM. G) CR strain cultures. After the organisms were inactivated by formalin, light mineral oil adjuvant with emulsifiers were added to the bacterial suspensions. The nine batches of bacterin were produced and then used to inoculate chickens for demonstration of efficacy and safety. The results showed: 1. Both bacterins were prepared with concentrated antigens by centrifugation and filtration methods are safety and efficacy in chicken, and there are no difference between them. 2. The birds vaccinated with 1:5 concentration bacterin can produce a good immunity due to more densely MG antigen concentration in the bacterin. But non-concentrated MG culture bacterin could not protect birds vaccinated from virulent challenge. There was not significantly different from those of non-immunized chickens in air-sac lesions. All the chickens did not develop of any untoward reactions after vaccinated with two doses bacterin per chick. The antibody could last more than 10 months in chickens. The M. G recovery rate from chicken tra-cheal was much lower in vaccinated birds than uninoculated control at 14 days post-challenge.
