【摘要】 根据发表的AEV序列设计一对引物,利用反转录-聚合酶链式反应(RT-PCR)技术,在体外从AEV分离株L2Z株感染的SPF鸡胚病变组织中扩增出VPO基因。将VP0基因克隆到质粒pBssK载体的Kpn Ⅰ酶切位点上,构建成含有VP0基因的重组质粒VP0-pBssK。进一步序列分析表明,该基因长726bp,编码242个氨基酸,与AEV1143株的核苷酸和氨基酸的同源性分别为95.2%和97.5%。与小RNA病毒科其它病毒属相应基因的氨基酸同源性比较发现,该病毒与人甲肝病毒同源性最高,在系统进化树上,与甲肝病毒之间距离最近,表明AEV与甲肝病毒属之间亲缘关系密切,给AEV病毒的确切分类提供了重要的实验数据。更多还原

【Abstract】 A pair of primers for avian encephalomyelitis virus (AEV) isolate L2Z was designed according to published AEV genome. With use of reverse transcription-polymerase chain reaction (RT-PCR), fragment VPO was in vitro amplified from Special-Pathogen-Free (SPF) chick embryo tissue after inoculation of AEV isolate L2Z. The fragment VPO was inserted into vector pBssK to obtain the recombinant plasmid VPO-pBssK with the length of 3. 7kb. VPO gene sequence was determined with the length of 726bp, and its deduced acid sequence was 242aa. By comparison with other published viruses of Picornaviridae, the nucleotide sequence and deduced amino sequence of VPO of the strain were 95. 2% and 97. 5% homologies with the correspondent sequences of AEV strain 1143 Respectivly and its nucleotide sequence and deduced amino sequence were 50. 1% and 50. 8% identities with hepatitis A virus strain HAS-15 respectively, and showing lower identities with those of any other picornviruses. This showed that AEV will be tentatively reassigned to a genus other than the enterovirus genus within the virus family Picornaviridae.更多还原
