薄荷品种间遗传关系分析与植物学性状和精油成分差异

【摘要】 为明确薄荷品种间亲缘关系和表现型、化学型的不同,本研究首先利用 ISSR(Inter-simple sequence repeat)分子标记技术对11个薄荷品种进行了遗传多样性和相似性的分析,并根据聚类分析的结果将所测试薄荷归为4大类。在此基础上从每类中分别选取在上海地区引种成功的黑胡椒薄荷(Mentha ×pipefita cv.)、薰衣草薄荷(Mentha pipefita cv.’lavandula’)、菠萝薄荷(Metha suaveolens cv.‘variegata’)以及留兰香(Mentha spicata)(化学型Ⅱ),对其进行植物学性状和精油成分的分析比较。结果显示,四种薄荷的含油率和精油成分差异显著:黑胡椒薄荷和薰衣草薄荷的精油含有率较高,菠萝薄荷和留兰香的精油含有率低。黑胡椒薄荷精油的主要成分为薄荷呋喃27.49%、薄荷醇27.47%以及薄荷酮18.76%等,薰衣草薄荷精油中芳樟醇51.11%和乙酸芳樟酯13.58%是其主要成分,菠萝薄荷精油中氧化胡椒烯酮的含量超过80%,留兰香精油的主要成分为氧化胡椒烯酮39.66%和柠檬烯20.49%,从而证明 ISSR 分子标记技术能够从根本上表达遗传关系的远近,植物学性状和精油成分的分析能够进一步阐明品种的表现性状与化学型的差异。

【Abstract】 In order to make clear about the heredity relations between varieties of Mentha and the difference of phenotype and chemical,eleven varieties of Mentha were analyzed by ISSR molecular marker technology.Clustering analysis showed that the 11 varieties of Mentha could be distinctively classified into 4 groups.Then we selected four varieties of Mentha separately from each group cultivated successfully in Shanghai,blackpepper mint(Mentha×piperita cv.),lavender mint (Mentha piperita cv.’lavandula’),pineapple mint (Metha suaveolens cv.’variegata’)and spear mint(Mentha spicata)(chemotype Ⅱ).The botanical characters and the composition of essential oil from them were analyzed and compared.The result showed that there was significant difference among the four varieties of Mentha;the content of essential oil of blackpepper mint and lavender mint is higher than that of pineapple mint and spear mint;menthofuran,menthol and menthone are the three main components of the essential oil from blackpepper mint,with 27.49%,27.47% and 18.76% respectively;the major components of essential oil of lavender mint were linalool and linalyl acetate, accounting for 51.11% and 13.58% respectively;the content of piperitenone oxide in the essential oil of pineapple mint exceeded 80%;in spear mint,the major components are piperitenone oxide and limonene, with 39.66% and 20.49% respectively.Thus proved that ISSR molecular marker technology can fundamentally express the heredity relations,botany character comparison and composition of essential oil analysis can further expound the variety performance character and the chemical difference.
