积雪草甙、羟基积雪草甙对成纤维细胞Ⅰ、Ⅲ型胶原合成的影响

【摘要】 目的:探讨积雪草甙和羟基积雪草甙对体外培养成纤维细胞(Human Skin Fibroblast, HSFb)Ⅰ、Ⅲ型胶原合成的影响。方法:通过胶原酶消化法体外培养HSFb,在不同剂量积雪草甙和羟基积雪草甙作用后,应用放射免疫的方法测定实验组和空白对照组细胞上清中Ⅰ型和Ⅲ型胶原的前胶原氨基端肽原((Procollagen Ⅰ of aminoterminal propeptide,PⅠNP;Procollagen Ⅲ amino-end propeptide,PⅢNP)含量。结果:(1)与空白对照组相比,积雪草甙和羟基积雪草甙可有效刺激体外培养HSFb PⅠNP和PⅢNP的分泌;(2)积雪草甙和羟基积雪草甙刺激靶细胞PⅠNP和PⅢNP分泌的生物学作用呈现明显的量效关系;(3)在10~150μg/ml剂量范围内,Ad对HSFb刺激PⅠNP的分泌显示出比Md更强的生物活性(p<0.01)。结论:Ad和Md对促进体外培养HSFbⅠ、Ⅲ型的胶原合成有明显的生物活性。

【Abstract】 Aim: To study the effects of asiaticoside and madcassiaside on the type Ⅰ and type Ⅲ collagen synthesis in cultuted human skin fibroblast in vitro. Method: The different dose asiaticoside and madecassiside were used in cultured fibroblast in vitro that obtained by collagenase digestion culture technology. The protein content of PⅠNP 和 PⅢNP in the culture medium of treated groups and control groups were determined by radioimmunoassay. Results: (1) Compared with control groups, the results shown an obvious stimulation of PⅠNP and PⅢNP protein secretion in the treated groups of Asiaticoside and madcassiaside. (2) The pharmacological action of asiaticoside and madecassiside presented a dose-dependent manner. (3) Asiaticoside shown a stronger biologyal activity than madecassiside for the PⅠNP excretion in fibroblasts culture medium at the concentration range of 10μg/ml and 150μg/ml (P<0.01) . Conclusion: Both asiaticoside and madcassiaside exhibit significant biology activities on stimulation of collagen type Ⅰ and Ⅲ synthesis in cultured fibroblasts in vitro.
