积雪草甙、羟基积雪草甙对体外培养皮肤细胞生物学特征的影响

【摘要】 目的:探讨积雪草甙(Asiaticoside,Ad)、羟基积雪草甙(Madeassoside,Md)对体外培养细胞生物学特征的影响。方法:通过体外培养角质形成细胞和成纤维细胞, 应用XTT方法和流式细胞仪比较研究积雪草甙和羟基积雪草甙对靶细胞增殖和细胞周期的影响。结果:积雪草甙和羟基积雪草甙在高达200μg/ml浓度时未见任何细胞毒性;对角质形成细胞和成纤维细胞有明显促进增值和 DNA合成作用(p<0.05);积雪草甙比羟基积雪草甙有更强的生物活性(p<0.05)。结论:积雪草甙和羟基积雪草甙对人皮肤角质细胞和成纤维细胞均具有明显的生物活性。

【Abstract】 Aim: The effect of asiaticoside and madcassiaside biology characteristic on the cultuted skin cells was demonstrated. Method: The test substances extracted from Centella asiatica were applied in this study. The influence action of asiaticoside and madcassiaside were determined on proliferation and DNA synthesis of cultured keratinocyte and fibroblast by XTT method and flow cytometry. Results: Asiaticoside and madcassiaside haven’t any cytotoxicity at the concentration of 200μg/ml. The increases of cells proliferation and DNA content were showed in the keratinocyte and fibroblast treated with both asiaticoside and madecassiaside (p<0.05). Meanwhile, asiaticoside exerted a stronger biology activity than madcassiaside at different doses in the keratinocytes and fibroblasts (p<0.05) . Conclusion: Both asiaticoside and madcassiaside exhibit a significant biology activity in the keratinocyte and fibroblast.
