幽门螺杆菌感染的胃黏膜病变组织中硒与PCNA表达的关系

【摘要】 目的研究幽门螺杆菌(Helicobacter pylori,HP)感染的胃黏膜病变中组织硒含量与PCNA表达的关系。方法应用快速脲酶试验,结合Warthin-Starry银染色确定胃黏膜病变标本的HP感染;应用免疫组化染色法检测PCNA的表达:应用MK光钎压力密闭微波溶解炉及RF-1501荧光分光光度计检测组织硒含量。结果各种病变胃黏膜组织硒含量除DYS与GC无显著性差异外(P>0.05),其余各组均有显著性差异(P<0.05),在CSG中硒含量显著性升高,而随病变程度加重,硒含量相对降低(P<0.05),但均高于正常组(P<0.05);各组胃黏膜病变中,PCNA表达HP阳性标本(P<0.01);HP感染胃黏膜组织硒含量与PCNA表达呈负相关(rs=-0.9898,P<0.05)。结论HP感染直接或间接地促进PCNA的表达;组织硒含量随病变程度加重相对降低,但高于正常组。HP感染的胃黏膜组织硒含量明显增高可能是保护性地对抗增加的活性氧的反应,亦可能是与某些癌基因表达之间存在着联系,直接或间接的抑制了癌基因表达,从而阻抑由HP感染而诱发的细胞恶性转化。

【Abstract】 Objective:To study the relationship between expression of PCNA and selenium in gastric mucous membrane lesions with HP infection methods.Gastric mucous membrane lesions with HP infection were examined by both of Warthin-Starry and RUT,expression of PCNA was investigated by using immunohistochemistry,selenium level in gastric mucous membrane was determined by MK microwave oven and RF-1501 spectrofluorometer.Ruselts:Selenium level in gastric mucous membrane lesions was significantly distinct in different groups except DYS and GC (P<0.05),Selenium level in CSG group with HP infection was significantly increased and declined with the deterioration of disease but was also higher than that in normal tissue (P<0.05).Expression of PCNA was higher in HP+ group than in HP" (P<0.01).Selenium and PCNA in HP infected diseases had a negative correlation (rs=-0.9898,P<0.05).Conclusions:Infection of HP directly or indirectly promotes the expression of PCNA.Concentration of selenium decline with the deterioration of diseases but is still higher than normal tissue.It shows that high concentration selenium in gastric mucous membrane infected by HP may not only be a protective response against increased oxidative stress,but also may inhibit the expression of different oncogenes and effectively prevent body from HP infection and deterioration of diseases.
