Caspase-3、8、9mRNA在实验性淋巴滞留性脑水肿大鼠模型中的表达研究

【摘要】 目的:探讨caspase-3、8、9mRNA在淋巴滞留性脑病大鼠实验动物模型中的表达情况及其与神经元凋亡之间的关系。方法:成年雄性Wistar大鼠,随机分为手术组、假手术组和对照组,手术组建立淋巴滞留性脑病实验动物模型,在损伤后不同时间点(1d、3d、5d、7d、14d)取颞顶皮层、海马区脑组织,用半定量RT-PCR方法观察淋巴滞留性脑水肿大鼠损伤后caspase-3、8、9mRNA的表达情况。用假手术组、对照组观察并对比上述指标。结果:Caspase-3 mRNA在淋巴阻塞后3天内颞顶皮层、海马中均表达上调,以第一天表达最高。Caspase-8 mRNA表达亦增高,但Caspase-9 mRNA表达无明显变化。结论:提示在淋巴阻塞性脑水肿实验动物模型中神经元凋亡的发生与Caspase-3、8的激活有关,并且神经元凋亡的发生可能绕过了线粒体途径。
【Abstract】 Objective: To investigate the expression of caspase 3、8、9 mRNA in rat model of lymphostatic encephalopathy and its relationship between neuron apoptosis and lymphostatic encephalopathy.Methods: 56 male adult Wistar rat were randomly divided into 3 groups:operation group, sham-operation group and control。We use operation group to establish the rat model of lymphostatic encephalopathy, after different intervals (1d, 3d, 5d, 7d, 14d),animals were sacrificed, temporoparietal cortex and hippocampal tissue were isolated and stored in -80℃ for late use..Semi-quantative RT-PCR method was used to explore the expression of caspase mRNA.Results:Caspase 3 mRNA is up-regulated in temporoparietal cortex and hippocampus within 3 days,the highest expression occurs in the first day after operation.The same condition occurs in caspase 8.But caspase 9 mRNA expression has no significant change.Conclusion:In lymphostatic encephalopathy rat model,neuron apoptosis is higuly related to the activation of caspase 3、8 and is lowly related to the mitochondrial way.
