四氧嘧啶糖尿病大鼠主动脉元素含量与收缩反应的相关性

【摘要】 目的:了解糖尿病时主动脉12种元素含量变化与其收缩反应的相关性。方法:用原子吸收光谱法测定四氧嘧啶糖尿病4周大鼠(n=12)及对照大鼠(n=12)主动脉组织中钙(Ca)、钼(Mo)、锂(Li)、锰(Mn)、镉(Cd)、镁(Mg)、铅(Pd)、钴(Co)、铁(Fe)、铜(Cu)、锌(Zn)及铬(Cr)的含量;用张力换能器和生理记录仪测定大鼠主动脉对10~-~6mmol/L苯肾上腺素收缩反应的张力;并对元素含量与主动脉环收缩反应张力之间的相关性进行分析。结果:糖尿病4周大鼠主动脉Ca、Pd、Cu、Fe及Cr含量显著高于对照(p<0.001)。糖尿病大鼠主动脉环对苯肾上腺素收缩反应的张力显著高于对照组(P<0.01);糖尿病大鼠主动脉Ca含量与收缩反应的张力呈显著正相关(r=0.746,p<0.01)。结论:糖尿病时主动脉存在Ca、Pb、Cu、Fe及Cr代谢异常;糖尿病时主动脉收缩反应增强与其Ca含量增加有关。主动脉Ca含量增加可能导致收缩反应增强。

【Abstract】 To find out change of elements content and relationship with contractile tension in aorta of diabetic condition. METHOD: Calcium(Ca), molybdenum(Mo), lithium(Li), manganese(Mn), cadmium(Cd), magnesium(Mg), lead(Pd), cobalt(Co), iron(Fe), copper(Cu), zinc(Zn) and chromium(Cr) content of aorta was determined by atomic absorption spectroscopy in 4 weeks alloxan-induced diabetic rats and control rats. Tension of contractile response to 10-6 M phenylephrine in aortic ring from rats was determined with a tension-power transformer and a physiological recorder. Relationship of element content in aorta with tension of aortic rings was analyzed in diabetic rats. RESULTS: Aortic Ca, Pd, Fe, Cu and Cr content in 4 weeks diabetic rats was significantly higher than in control rats (p<0.001). Tension of contractile response to 10-6 M phenylephrine in aortic rings from diabetic rats were significantly higher than from control rats (p<0.01), Calcium content of aorta were positive relationship with tension of contractile response in aortic rings from diabetic rats (r=0.746, p<0.01). CONCLUSION: The result suggests that there is metabolic abnormality of Ca, Pd, Fe, Cu and Cr, and relationship of enhancement of contractile response with increase Ca content in aorta from diabetic rat.
