盐芥ThHKT1基因的克隆

【摘要】 <正>我们以盐芥为材料,依据拟南芥AtHKT1基因的序列设计引物,扩增出盐芥中ThHKT1基因的部分序列。然后使用快速扩增(RACE)方法,从盐芥中克隆到ThHKT1基因cDNA序列。该序列全长1817bp,推测编码区长度为1503bp,编码500个氨基酸,等电点为8.82。其编码序列和氨基酸
【Abstract】 A gene fragment was amplified by a pair of primers designed from Arabidopsis AtHKT1 gene from Thellungiella halophila. With the ThHKT1 gene fragment, the full cDNA sequence of ThHKT1 gene which was 1817bp long was obtained by RACE. Further analyses on the ThHKT1 gene indicated that an open reading frame (ORF) of 1503bp in this gene encodes 500 amino acids. The codon sequence and amino acid sequence of the ThHKT1 gene shared 82.8% and 77.3% identity with those of AtHKT1 gene respectively.
