应用实时荧光PCR方法检测转基因植物产品
Detection of Genetically Modified Plant Products with Real-time Fluorescent PCR Method

【摘要】 本研究根据商品化转基因植物信息以及基因序列,设计了CaMV 35S启动子、nos终止子、npt Ⅱ、pat、bar和FMV 35S启动子的特异引物及TaqMan荧光探针用于转基因植物产品的定性筛选检测,以核糖体18S rRNA基因作为内参照以监控DNA的提取质量。结果表明,所用的引物和探针具有良好的特异性,18S rRNA可作为植物DNA提取的质控基因。实时荧光PCR技术在检测速度、防止假阳性和灵敏度等方面均优于常规的PCR检测方法。
【Abstract】 A real-time fluorescent PCR method was established to detect genetically modified plant products (GMPs). Seven TaqMan probes were designed for qualitative screening detection of GMPs and ribosomal 18S rRNA probe was used to measure the DNA quality in this method. The results showed that all the probes have perfect specificity and 18S rRNA could be used as the reference gene, which indicated that the method was suitable for the screening detection of GMPs. In addition, the real-time PCR was more specific and sensitive than normal PCR.
