双孢蘑菇2796菌株液体发酵培养基的优化

【摘要】 通过单因子试验,对双孢蘑菇2796菌株发酵培养基进行优化,确定最佳摇瓶培养基的碳源为玉米淀粉、氮源为豆饼粉、无机盐为K2HPO4、MgSO4·7H2O、NaCl和CaCl2。最佳发酵条件为25℃,起始pH6.5, 采用SAS软件中的Plackett-Burman设计从八个考察因素选出玉米淀粉、豆饼粉和MgSO4·7H2O为影响实验的三个重要因素;通过响应面分析法建立模型:预测最大响应值为2.52g/100mL,经摇瓶发酵实验证实此点的实测值为2.54g/mL,证明模型是有效的,并且存在极大值点;并得到优化后的发酵培养基配方。

【Abstract】 The optimal fermentation conditions were worked out through single factor test. The experiments show that cornstarch .soybean cake are the optimum carbon and nitrogen sources. K2HPO4, MgSO4 7H2O, NaCl and CaCl2are inorganic salt. The optimum fermentation conditions are initial pH 6.5, fermentation temperature 25℃. By using Plackett-Burman design by SAS software, cornstarch .soybean cake and MgSO4·7H2O were chosen as the major components from eight variables. A regression model equation was obtained after Response Surface Analysis(RSA) performed. The optimum concentrations for the maximum dry weight, which is 2.52g/100mL.The model was validated experimentally by the maximum production of 2.54g/mL.
