灵芝子实体和孢子中总三萜含量的比色法测定

【摘要】 本文报道了一种灵芝(Ganoderma lucidum)子实体和孢子中总三萜含量的比色测定方法。灵芝总三萜用乙醇提取,与香草醛及高氯酸在加热条件下发生显色反应,最大吸光值在560nm左右,与齐墩果酸标准品反应后基本一致。以齐墩果酸作标准曲线,检测线性范围是9-75μg/ml,反应物至少在1小时内稳定。用该法测得灵芝子实体和孢子中总三萜含量分别为1.07%和0.57%,回收率为98.2%。
【Abstract】 A colorimetric method was developed for the detection of total triterpene contents in the fruiting bodies and spores of Ganoderma lucidum (GL). The total amount of triterpenes was successfully quantified when the ethano-licethanol extracts of CL were reacted with vanillin and perchloric acid under heating condition. The developed color showed a maximum absorption at 560nm, which was proximate to that of the standard oleanolic acid used as a standard, and the color was stable for at least one hour. The standard curve was linear in a range from 9 to 75μg/ml, and the total triterpene contents in fruiting bodies and spores were calculated as 1.07% and 0.57%, respectively using the oleanolic standard curve, which was linearily measured in a range from 9 to 75μg/ml . The recovery rate was calculated as 98.2%.
