胶孢炭疽菌的营养体亲和性

【摘要】 采用硝酸盐利用缺陷型突变体互补测试法, 时28个胶孢炭疽菌株(Colletotrichum gleosporiodes)的营养体亲和性进行了初步研究。通过WAC培养基进行抗氯酸盐诱变,获得4511个抗氯酸盐突变体,在CDA培养基上有1323个(占29.3%)被鉴定为nit突变体,其中 nit1型突变体占73.3%,NitM型突变体为7.7%,nit3型突变体为10.4%,nitD型突变体为8.6%。28个菌株中, 22个(78.6%)获得了nit突变体,被划分为7个VCGs: 13个菌株划为VCG1,属优势类群;3个菌株划为VCG2; 6个菌株没有和其它任何菌株产生亲和反应,仅仅是自身亲和,分别各为一个类群。结果表明C.gloeosporioides 群体具有丰富的VCG多样性,但与寄主种类、地理来源没有明显的相关性。

【Abstract】 With nitrate non- utilizing (nit) mutants complementary test, 28 Colletotrichum gleosporiodes isolates collected from different regions and different hosts were used to test vegetative compatibility. nit mutants were generated on WAC and identified on CD A. A total of 1323 nit mutants were selected from 4511 sectors and used for the further study. Of these nit mutants, 73.3% were identified as nit 1, 7.7% as NitM, 10.4% as nit3, 8.6% as nit D (unable to utilize nitrate, nitrite and hypoxanthine) . Among the twenty - eight tested isolates, twenty - two isolates (78.6% ) generated nit mutants were designated eight VCGs. 13 isolates were assigned to VCG1, while 3 isolates to VCG2. The remained 6 isolates showed only self- compatibility and be assigned to six different VCG. These results suggested that Col-letotrichum gleosporiodes exist abundant VCG diversity that had no close relationship with both host and geographical origin.
