新疆平盘石耳地衣的组织培养
Study on the Tissue Culture of Umbilicaria in Xinjiang

【摘要】 地衣(Lichen)是菌藻共生体,随着分子生物学技术以及细胞工程的发展,地衣的经济价值日益显露出来,引起了各方面的兴趣,在抗癌药物、生物制剂、香料工业等诸多方面有很大的发展潜能,大多数地衣特有的多糖具有高度的抗癌活性和增强免疫的功能,尤其是石耳属地衣所含有的多糖物质的抗癌活性已达到90%以上,在生态环境方面,地衣常被作为监测大气污染的指示生物,因此,我们对能否在不破坏生态环境的条件下对其进行开发利用,能否用人工方法实现新疆石耳属地衣的组织培养进行初步探索。本文介绍了根据已知的真菌、藻类和蓝细菌培养的培养方法的基础上,对新疆石耳属地衣(Umbilicariahaffm)平盘石耳(V.leiocarpa Ds)在地衣型真菌培养基,共球藻有机营养培养基和蓝细菌培养基上对其进行的组织培养结果,结果表明:此次实验我们采用的五种培养基中麦芽汁—酵母膏培养是平盘石耳地衣型真菌较为理想的培养基,而共球藻有机营养培养基对其共生藻生长也是比较适合的,在地衣型真菌培养基即麦芽汁—酵母膏培养基上的组织表面出现了细小的子束盘(nang),并数目较多,而在地衣型共球藻有机培养基上,组织块表面的褶皱顶端及组织块周围出现有白色生长物的现象,从这些结果看,我们认为很可能是因为地衣是真菌和藻类的共生体,所以恰好适合在于这以上培养基中生长,根据以上实验结果,成功的建立了新疆平盘石耳地衣组织培养的初步基础模型。
【Abstract】 Abstarct: As symbiotic organisms of algae and fungi. Lichens and their economic value is becoming more andmore important along with the development of the research techniques in molecular biology and cell engineeringand have been brought to human being’s attention. In this paper,the results of tissue culture of Umbilicaria V.leiocarpa Ds species from Xinjiang in three different kinds of culture medium: lichenized fungi culture medium.phycobiont organic nutrition culture medium and blue-green algae culture medium are reported. We foundthat a-mong the five kinds of culture medium the maltose yeast culture medium is a kind of most suitable one for thetissue culture of Umbilicaria lichen; phycobiont organic nutrition culture medium is better for the tissue cultureof phycobiont; on the lichenized fungi culture medium maltose yeast culture medium,some tiny nangs with greatnumbers had been appeared; a kind of white coloured grown object had been found round the edge of the cul-tured tissues and on the top parts of wrinkles on the surface of the cultured tissues. According to these resultswe concluded that lichen tissue is suitable for growing these culture medium,because lichen is a symbiotic offungiand algae. On the basis of this experiment we have successfully established the preliminary basic model ofthe tissue culture of Umbilicaria v. leiocarpa Ds lichen from Xinjiang.
