甘蓝型油菜磷营养高效种质的筛选与QTL定位

【摘要】 对243个甘蓝型油菜品种苗期和全生育期缺磷胁迫反应的适应性差异进行筛选,获得磷高效品种97081和磷低效品种97009。在缺磷条件下,磷低效品种97009苗期地上部干重与正常磷处理下的比值为0.417,籽粒产量的比值为0.160,而高效品种97081分别为0.755和0.348。高效品种在缺磷的条件下地上部和根系吸收累积的磷大于低效品种,且根部的磷输送到地上部的比例较大。以一个F2:3家系为作图群体,应用SSR和AFLP分子标记,构建了一个包含23个连锁群的遗传连锁图谱。采用QTL分析软件WinCarter2.0对缺磷的籽粒产量(DSY)及其磷效率系数(PEC)进行QTL扫描,共检测到4个与之显著相关的QTL,DSY1、DSY2、DSY3和PEC1分别位于第2、11、12和21连锁群上,单位点解释的表型变异方差为19.4%~41.4%。研究结果表明油莱磷营养高效是受多个基因控制,具有复杂的生理或分子机制。

【Abstract】 Phosphorus (P) is an essential mineral nutrient for plant growth, and Brassica napus is an important oil crops which is sensitive to P deficiency. In this study, a P-efficient cultivar ’97081’ and a P-inefficient cultivar ’97009’ were obtained by screening 243 cultivars of Brassica naptu at seedling stage and whole growth period. Ratio of shoot dry weight at P deficiency treatment to that at P normal treatment in seedling stage for the P-inefficient cultivar was 0.417, and the ratio of seed yield for P deficient and normal treatments was 0.160. However, the two ratioes for the P-efficient cultivar were 0.755 and 0.348, respectively, which was higher than P-inefficient cultivar. P efficient cultivar ’97081’ accumulated more P than P-inefficient cultivar ’97009’ both in root and shoot at seedling stage. A genetic linkage map with 23 linkage groups was constructed using SSR and AFLP markers in a F2:3 population, and then genome-wide QTL analyses with WinCarter2.0 software detected 4 QTLs which significant related to seed yield (DSY) at phosphorus deficiency (DSY1, DSY2, DSY3) and phosphorus efficiency coefficient (PEC1) . These QTLs located on linkage groups 2, 11, 12 and 21, respectively and explained phenotypic variance from 19.4 % to 41.4 % . This suggested that multigenes are involved in uptake and utilization of phosphorus in Brassica napu, which respond to phosphorus deficiency stress and make sure more biomass and seed yield under the condition of phosphorus deficiency.
