测定原花青素平均聚合度的一种新方法

【摘要】 建立了一种测定葡萄籽提取物中原花青素平均聚合度的新方法。该方法首先以甲醇为溶剂,以传统香草醛法测定原花青素的质量,再以乙酸为溶剂,使香草醛只与原花青素末端黄烷-3- 醇反应来测定原花青素物质的量,然后通过对比获得原花青素的平均聚合度。优化后测定原花青素样品物质的量的线性浓度范围为0.028-0.345 μmol/mL,检测限为2.97×10-4 μ mol/mL,加标回收率为95.2%-99.6%,精密度(RSD)为0.78%-0.98%。通过该法测定的样品平均聚合度与实际值接近,具有简单、快速、准确、灵敏等优点。

【Abstract】 A new method was developed for determination of the average degree of polymerization (DP)of proanthocyanidins in extract of grape seeds.In the conventional vanillin assay,methanol is the solvent for determination of proanthocyanidins content.On the other hand,when the reaction was carried out in glacial acetic acid,only the terminal flvan-3-ol unit reacts with vanillin and thus determined the concentration of proanthocyanidins molecules.By using of these two reactions,the average degree of polymerization of proanthocyanidins can be determined.The linear range of determination was 0.028-0.345 μmol/mL,and the detection limit was 2.97×10-4 μmol/mL, respectively.The recovery was 95.2%~99.6% and the relative standard deviation was 0.78%~0.98%. The method is easy,fast,accurate,sensitive and has been applied to the samples determination and the results agree well with literature values.
