The anti-epileptic effect of BmK IT2 on rat seizures induced by pentylenetetrazol

【摘要】 <正>The anti-epileptic effect of BmK IT2. a sodium channel-specific modulator purified from Chinese scorpion Buthus martensi Karsch, on seizures induced by pentylenetetrazol (PTZ) in rats was investigated. The unilateral injection of BmK IT2 (at doses of 0.01. 0.05, 0.1, 0.5 and 1 mg) into the CA1 region of the hippocampus, could inhibit the seizures induced by intraperitoneal injection of PTZ. The suitable dose of BmK IT2 was determined between 0.1 and 0.5 mg. BmK IT2 could significantly prolong the latent period of epileptic seizures, decrease the stage of seizures, and shorten the duration of epileptiform discharges. The results showed that the strong epileptic behaviors and discharges in hippocampus induced by PTZ could be inhibited by intrahippocampal pretreatment of BmK IT2. The significant anti-epileptic effect of BmK IT2 found in this study suggests that BmK IT2 might be useful as a potential drug in the therapy of epilepsy.
