骨髓间充质干细胞向雪旺细胞分化的体内、外实验研究

【摘要】 目的研究SD 大鼠的骨髓间充质干细胞在体内、外条件下向周围神经的雪旺细胞分化的可行性。方法从SD 大鼠的股骨和胫骨获得间充质干细胞,利用间充质干细胞贴壁生长的特性培养纯化,传代扩增。复合诱导因子(Beta-mercaptoethanol,retinoic acid,forskolin,basic-FGF,PDGF,heregulin)体外诱导间充质干细胞分化。免疫细胞化学(p75,S-100,GFAP)鉴定体外的诱导结果。同时将PKH67标记的间充质干细胞移植入损伤的周围神经内,两周后组织切片免疫荧光染色,激光共聚焦定位移植的细胞去向。结果诱导后的间充质干细胞形态类似雪旺细胞,免疫荧光鉴定骨髓间充质干细胞具有雪旺细胞性质,表达雪旺细胞的表面标志(GFAP,S100和P75)。周围神经组织切片免疫荧光染色后共聚焦定位的间充质干细胞表达雪旺细胞的表面标志(GFAP,S100和P75)。结论骨髓间充质干细胞在体内、外都可以分化为具有雪旺细胞性质的细胞,表达GFAP,S100和P75。骨髓间充质干细胞有可能替代雪旺细胞促进周围神经的再生。
【Abstract】 Objective To explore the possibilities of bone marrow stromalcells(MSCs)to take on Schwann cell’s phenotype in vitro and in vivo in SDrats.Methods MSCs were gotten from the tibia and femur bone marrow andcultured by their adherence characteristics to culture flask.Beta-mercaptoethanol followed by retinoic acid,forskolin,basic-FGF,PDGFand heregulin were added to induce MSCs’ differentiation.Schwann Cellmarkers,p75,S-100 and GFAP were used to discriminate the induced MSCs’properties by immunofluorescent staining.PKH-67-labeld MSCs weretransplanted into the mechanically injured rat sciatic nerve and laser confocal microscopy was used to localize the PKH67 labeled MSCs in injuredsciatic nerve two weeks after operation.Fluorescence PKH67 attenuation rulewas evaluated by flow cytometry in vitro.Results MSCs changed morphologicallyinto cells resembling primary cultured Schwann cells after induction in vitro.In vivo,a large number of MSCs were cumulated within the layer of epineuriaaround the injured nerve and expressed Schwann Cell markers,p75,S-100,andGFAP.Conclusion The seresults suggested that MSCs have the capabilities tosupport nerve fiber regeneration and re-myelination by taking on Schwann cellfunction,and are potentially useful as the possible substitutable cell forartificial nerve conduits to promote nerve regeneration.
