宫内感染致早产鼠脑损伤动物模型建立的实验研究

【摘要】 目的探索宫内感染致早产仔鼠脑损伤动物模型的制备方法。方法通过给予孕16、17天大鼠连续两天腹腔注射脂多糖(LPS)350 μg/kg,制备宫内感染致早产仔鼠脑损伤动物模型。光镜与电镜下观察7、30日龄早产仔鼠脑组织变化,于早产鼠生后30天进行行为学检测。结果有6只大鼠提前分娩,共获活早产仔鼠26只。与随机选取的足月产仔鼠比较,早产鼠出生后颜色青紫、活动少、体重低;病理结果显示早产动物模型脑损伤部位在白质,系胶质细胞、髓鞘的损伤。早产鼠感觉运动功能较足月产仔鼠明显减退。结论孕鼠腹腔注射脂多糖可成功诱发早产动物模型,该早产动物模型存在脑损伤,其病理改变以白质损伤为主。
【Abstract】 Objective: To establish a new preterm rat model with brain injury induced by intrauterine infection. Method: Pregnant rats were consecutively injected with LPS (350μg/kg) or saline on gestation d 16 and 17 intraperitoneally. The interval from infusion to delivery for each group was recorded, along with the number of live born pups and their birth weight Preterm pups and normal term pups were divided into groups: d 7 after labor and d 30, respectively. The brain damage of the preterm rats was observed by optical and electron microscopes. The behavioral abilities of premature pups were tested till d 30. Results: There were 26 live preterm pups delivered by 6 LPS -treated rats. The birth weight of the pups reduced compared with the saline controls. And there were motor reduction of preterm pups. There was intrauterine infection of LPS group rats. The behavioral abilities of premature pups were much lower than those of normal control group. There was white matter damage of animal modal. Conclusion: Preterm delivery can be induced by LPS. A new animal model with brain injury was established by LPS-induced preterm. The damage location of the animal model was white matter.
