核因子kappaB抑制剂DHMEQ对头颈鳞癌细胞体外增殖及侵袭的抑制作用
Growth and invasion inhibitory effect of DHMEQ (inhibitor of nuclear factor kappa B) on head and neck squamous cell carcinoma cell lines in vitro

【摘要】 目的:研究核因子kappa B(Nuclear factor kappa B,N F- κ B)抑制剂-脱氢环氧甲基醌霉素 (Dehydroxymethylepoxyquinomicin,DHMEQ)对头颈鳞癌细胞体外增殖及侵袭的抑制作用。方法:选择口腔鳞癌细胞株KB、鼻咽鳞癌细胞株YCU—N861及下咽鳞癌细胞株 YCU—H891为研究对象,应用WST-1快速比色法研究三种头颈鳞癌细胞株对DHMEQ的敏感性;应用细胞计数法检测 D H M E Q对三种头颈鳞癌细胞株体外增殖的抑制;应用 ELLISA细胞死亡检测试剂盒,来定量检测头颈鳞癌细胞的凋亡;应用invasion chamber试剂盒检测DHMEQ对头颈鳞癌细胞侵袭力的影响;应用western blot法检测经 D H M E Q处理后细胞中基质金属蛋白酶9(m a t r i x metalloproteinases 9,MMP-9)的表达水平。结果:三种头颈鳞癌细胞对DHMEQ均敏感,DHMEQ对三种细胞50%的生存抑制浓度(IC50),均接近20 μ g/ml; DHMEQ对三种头颈鳞癌细胞的体外增殖均有明显的抑制作用;随 DHMEQ用药浓度的增加,三种细胞的凋亡也随之增加、细胞侵袭受抑制;随DHMEQ用药时间延长,头颈鳞癌细胞中 MMP-9蛋白的表达呈下降趋势。结论:DHMEQ可诱导头颈鳞癌细胞凋亡,从而抑制头颈鳞癌细胞体外增殖;DHMEQ 还能抑制头颈鳞癌细胞的侵袭。
【Abstract】 Objective: To investigate the growth and invasion inhibitory effect of DHMEQ (inhibitor of nuclear factor kappa B) on three head and neck squamous cell carcinoma (HNSCC) cell lines in vitro. Methods: Three HNSCC cell lines (KB, an oral cavity squamous cell carcinoma ceil line. YCU-N861, a nasopharyngeal squamous cell carcinoma cell carcinoma cell line and YCU-H891, a hypopharyngeal squamous cell carcinoma cell line) were used in this study. The sensitivity of three cell lines to DHMEQ was determined by WST-1 assay. Growth inhibitory effect of DHMEQ on HNSCC cell lines was detected by cell counting. Apoptotic cell death was assessed and quantified using the Cell Death Detection ELISA kit Invasion chamber kit was used to detect the change of invasion ability of HNSCC after the use of DHMEQ. Western blot was used to detect expression changes of matrix metalloproteinases 9 (MMP-9 ) after DHMEQ administration. Results: DHMEQ showed similar growth inhibitory effects on three cell lines, with an IC50 of about 20 μ g/ml. Treatment with DHMEQ induced apoptosis in a dose-dependent manner accounting, at least in part, for the growth inhibition by DHMEQ. Treatment with DHMEQ decreased the invasion ability of HNSCC in a dose-dependent manner. The expression of MMP-9 pro- tein was down regulated after DHMEQ administration. Conclusion: By inhibiting NF- κ B, DHMEQ can inhibit the proliferation of HNSCC cells in vitro through increased apoptosis. It can also inhibit the invasion of HNSCC cells in vitro.
