树突细胞活化的特异性CTL对人胃癌裸鼠移植瘤的作用

【摘要】 目的:探讨负载胃癌细胞抗原的树突细胞(DC)诱导特异性细胞毒性T细胞(CTL)对人胃癌裸鼠移植瘤的抗肿瘤作用。方法:提取、扩增外周血DC和T淋巴细胞并鉴定。建立人胃癌裸鼠移植瘤模型,随机分为实验组和对照组,皮下注射胃癌细胞冻融抗原负载或非抗原负载D C诱导的 CTL,测量肿瘤大小、重量,光镜、电镜观察形态学变化, 流式细胞术检测肿瘤细胞G0/G1、S、G2/M期细胞比率及凋亡率。结果:与对照组相比,实验组瘤体的体积和重量明显减小(P<0.01),形态学观察可见大量坏死溶解细胞,且实验组瘤细胞G0/G1期细胞比率和凋亡率显著增多(P< 0.05或0.01),而G2/M期细胞比率显著降低(P<0.05)。结论:负载胃癌细胞抗原DC激活的CTL在体内对肿瘤具有强大的的特异性细胞毒作用。
【Abstract】 Objective: To investigate the anti-tumor effect of the cytotoxicity T lymphocytes(CTL) activated by dendritic cells(DC) loaded gastric cancer cells antigen on human gastric carcinoma in nude mice. Methods: DC and T cells were isolated from human peripheral blood and cultured in vitro. The human gastric carcinoma models were established and divided randomly into experiment and control group, which were injected CTL activated by DC loaded with frozen-thawed gastric cancer cells antigen or not. After treatment, all model mice were killed to measure the tumors’ volume and weight and to observe the morphological changes by light and electronic microscope. Apoptotic rate and tumor cells cycle were detected by FCM. Results: The tumors’ volume and weight of experiment group were significantly smaller than that of control group (P < 0.01). Compared with control group, the apoptotic rate and tumor cells in G0/G1 stage was greatly increased (P < 0.05 or 0.01), and tumor cells in G2/M stage were dramatically decreased (P < 0.05). Many necrotic tumor cells were seen in experiment group. Conclusion: CTL activated by DC loaded with frozen-thawed tumor antigen has effective cytotoxicity against gastric carcinoma in viro.
