【摘要】 目的:探讨Cks1在胃癌发生及其在Skp2调节 P27Kipl降解过程中的作用。方法:应用流式细胞术测定正常胃粘膜、胃粘膜异型增生组织及胃癌组织中Cksl、P27Kipl、 Skp2蛋白表达。结果:由正常胃粘膜、胃粘膜异型增生组织到胃癌Cks1、Skp2表达呈上升趋势(P<0．05),P27Kipl 表达呈下降趋势(P<0．05)。胃癌中Cksl、Skp2表达与 P27Kipl表达呈负相关(r=-0．649,P<0．05;r=-0．732,P <0．05);而Cks1蛋白表达与Skp2蛋白缺乏相关性p> 0．05)。胃癌中Cks1的表达与肿瘤分化程度相关(P<0．05), 而与肿瘤浸润深度、淋巴结转移及临床分期不相关 (P> 0．05)。结论:Cks1可能参与了胃癌的发生;胃癌中Cks1 可能参与了Skp2调节P27Kipl泛素化降解过程。更多还原

【Abstract】 Objective: To investigate the expression of Cksl protein and its relationship with the expression of Skp2, P27Kipl and tumor aggressiveness. Methods: Flow cytometry was used to detect the expression of Cksl, Skp2 and P27Kipll in 20 cases of normal gastric tissue and 20 cases of atypical hyperplasia of gastric mucous membrane as well as 54 cases of primary gastric cancer. Results: The expression of Cksl and Skp2 gradually increased from normal gastric tissue, atypical hyperplasia of gastric mucous membrane to gastric cancer (P < 0.05). while the expression of P27Kipl gradually decreased P < 0.05). A significant inverse relation was observed between the expression of Cksl or Skp2 and of P27Kipl (r=-0.649, P < 0.05; r=-0.732, P < 0.05 respectively). The expression of Cksl was significantly associated with loss of tumor differentiation. (P < 0.05). Conclusion: Cksl may play an important role in human gastric tumorigenesis. Cksl may be an essential accessory factor for Skp2 in proteasome-dependent destruction of P27Kipl.更多还原
