荧光定量PCR检测乳腺癌组织syk基因的表达及其临床病理意义

【摘要】 目的:探讨syk基因在人乳腺癌组织中及其乳腺癌细胞株中的表达及其生物学意义。方法:采用荧光定量PCR 方法和免疫组织化学分别检测乳腺癌细胞株M C F-7、 MDA-MB-231、30例人乳腺正常组织、瘤体组织中 sykmRNA、蛋白表达及其临床病理特征的关系。结果:荧光定量PCR检测syk mRNA的表达:人乳腺癌细胞株 MCF-7中sykK基因mRNA表达为阳性,定量为6.59× 105; MDA-MB-231的syk基因mRNA为阴性。30例乳腺癌组织sykmRNA表达阳性率为63.3%(19/30)和正常乳腺组织中sykmRNA表达阳性率为93.3%(28/30)。乳腺癌癌组织sykmRNA的平均水平为(6.17±0.065)×104,明显低于配对的癌旁乳腺组织(80.3±0.125)×104,二者差异有统计学意义(P<0.01)。免疫组织化学法检测SYK蛋白的表达:人乳腺癌细胞株MCF-7中SYK蛋白表达为阳性, MDA-MB-231的SYK蛋白为阴性。30例乳腺癌患者中,乳腺癌、正常组织SYK蛋白表达阳性率分别为60%(18/30), 90%(27/30),两者之间有显著的统计学差异(P<0.01)。5例发生远处转移的乳腺癌浸润性导管癌中,syk表达缺失率占 80%。syk的mRNA和蛋白表达的下调与乳腺癌临床TNM 分期、腋淋巴结转移显著相关(P<0.05),而与肿瘤的大小, 病例分级、ER和PR状况无关(P>0.05)。结论:Syk在乳腺癌癌组织中低表达或不表达,而在乳腺癌正常组织中高表达,SYK基因低表达或缺失与肿瘤的浸润和转移有关。荧光定量PCR定量分析乳腺癌、正常组织内syk mRNA, sykmRNA的表达可以抑制肿瘤的生长,可以早期诊断及鉴别诊断乳腺癌。
【Abstract】 Objective: To explore the expression of syk gene in human breast cell and cancer tissue and its significance. Methods: Real-time polymerase chain reaction and immunohistochemical methods were performed to evaluate the expression of syk in 30 pairs of human breast cancer tissue and its surrounding tissues and two types breast cancer cell line and clinkopathological features was analysis. Results: Positive expression of Syk protein in MCF-7. Negative expression of Syk protein in MD A-MB-231; Reduced expression level of syk gene mRNA and protein has been noted in breast carcinoma tissue compared to adjacent non-cancerous breast tissue (P<0.01). The lower expression levels in breast carcinoma tissue was related with clinicopathological features including lymph-node metastasis and TNM staging(P<0.05), but not with the histo-logical grade nor the estrogen and progesterone receptor status(P>0.05). In breast carcinoma tissues, the level of syk mRNA is (6.17 ± 0.0.065 ) × 104, It is lower than level of syk mRNA in the normal tissues (80.3 ± 0.125 ) × 104. Conclusion: The expression of syk gene mRNA and protein was reduced in human invasive breast carcinoma. Down-regulated expression of SYK in breast carcinoma might be assiociated with tumor invasion and metastasis.
